Objective:To detect serum proteomic patterns in lung cancer by serface enhanced laser desorption/ionization-time of flight-mass spectrometry (SELDI-TOF-MS) techniques, screen biomarkers of lung cancer and build diagnostic models in order to evaluate their clinical significance.Methods:SELDI-TOF-MS and CM-10protein chip were used to detect the serum proteomic patterns of29squama cancers,29adenocarcinomas,and8small cell lung cancers.including34geminate patient serums with lung cancer pre-surgery and post-surgery and50normal control subjects. BioMarker Wizard and Biomarker pattern system and SPSS softwares were used in combination to analyze the data and to develop diagnostic models.Results:Two protein peaks from a total of174were chosen as a biomarker pattern in the training set. Two protein peaks pattern(mass/charge ratio[m/z]6010,5330) was observed in the model that could used to distinguish lung cancer from non cancerous diseases. It yielded a sensitivity of98%and a specificity of96%.There are nine different protein peaks between pre-surgery and post-surgery. There are17difference Serum protein between Peripheral lung cancer and central type lung cancer,25difference Serum protein between small cell lung cancer and lung squamous carcinoma patients,15difference Serum protein between small cell lung cancer and lung adenocarcinoma patients,three difference Serum protein between lung squamous carcinoma and lung adenocarcinoma patients, five differences serum protein between high differentiation in patients with lung cancer and differentiation of patients with lung cancer,three differences in Serum protein between high differentiation of patients with lung cancer and low differentiation lung cancer patients, two difference Serum protein between differentiation of patients with lung cancer and low differentiation of patients with lung cancer,19difference Serum protein between T2patients with lung cancer and T3lung cancer patients,and eight different peaks between metastasis and non-metastasis.Conclusions:These results suggest that SELDI techniques can distinguish lung cancer patients from normal subjects with relatively high sensitivity and specificity, and the SELDI-TOF-MS is a effective tool for the screening of lung cancer...
