| Preface Germ free rabbit has no parasitic pathogen, which immunologic system is in quiescence, so the immune reaction of germ free animal is intensive. The germ free rabbit model has a wide usage in future. The overseas studies on germ free rabbits focus on pathogenic bacterium. Normative establishments of germ free rabbits are the domestic studies. There are not systematic studies on germ free rabbits.Objective The requirement of high-quailty laboratory animals is increasing as the developing of life science. The germ free rabbits were used widely on the basis of systematic studies on domestic germ free rabbits. My systematic studies on germ free rabbits may be helpful references in the usage of germ free animal models.Methods The rabbits were cleansed by cesarean section, and the young rabbits were fed with artificial milk by hand in germfree isolator. Some of young rabbits were given the normal intestinal bacteria, which then became SPF rabbits. Biological characteristics in germ free, SPF and conventional New Zealand Rabbits were compared. The hematological and urine parameters in germ free, SPF and conventional rabbits were detected using automatic analyzer. The organ coefficients were measured by electronic balance. The histological structures of organs were assessed by microscopic examination of hematoxylin eosin staining. Blood parameters, blood biochemical parameters, urine routines, organ coefficients and histological structures were compared.Results The survival rate of young rabbits is 67.7%. In the comparison, blood parameters, blood biochemical parameters, urine routines, organ coefficients and histological structures are different:1. The RBC, RBC, HGB, HCT, MCV, MCHC, MCH, RDW, NE, MO, EO, BA, NE#, LY#, MO#, EO# and BA# had significant difference in the 3 groups of rabbits(P< 0.05). The PLT, MPV and LY had not significant difference(P>0.05);2. The TP, ALT, GLO, A/G, ALP, GGT, GLU, CREA, UA, Ca and HDL-C had significant difference in the 3 groups of rabbits(P<0.05). The ALB, AST, BUN, P, CHO, TG, LDL-C, CK and LDH had not significant difference(P>0.05);3. The SG and pH had significant difference in the 3 groups of rabbits(P<0.05). The LEU, NIT, PRO, GLU, KET, UBG, BIL and ERY in gnotobiotic rabbits had not significant difference compared with those in conventional rabbits(P>0.05); 4. The coefficients of left submaxillary gland, right submaxillary gland, heart, lung, cholecyst, spleen, left adrenal gland, right adrenal gland, womb+ovary, left testis, length of stomach, procussus vermiformis, length of caecum, gross weight of caecum, dry weight of caecum, gross weight of stomach and dry weight of stomach had significant difference in the 3 groups of rabbits(P<0.05). The brain, liver, left kidney, right kidney, pancreas, right testis, width of stomach, length of small intestine, length of rectum and length of large intestine had not significant difference(P>0.05);5. The rabbits had histological changes, conventional rabbits have higher inflammation rate compared with those in germ free and SPF rabbits.Conclusion The germ free rabbit model was established successfully by normative methods, the technique is maneuverable. In the comparison, the biological characteristics in germ free, SPF and conventional New Zealand Rabbits were different. Different microbiological background significantly affects the biological characteristics in New Zealand rabbits. |
