Dung Beetle is a dried scorpio of Catharsius molossus Linnaeus in Scarabaeidae.It is salty in flavor and cold in nature, poisonous,and has the meridian distribution to hepar,gaster and large intestinal.Its actions include relieving convulsion,removing blood stasis, stopping spasm,detumescence,eliminating toxic material,opening bowels,antifebric and so on.On clinic application,it is used to treat convulsive epilepsy and vesania,constipation,blennorrhagia, malnutritional stagnation,caco-sore,kinds of cancers, splenohepatomegalia and so on.Its main habitats are Shan Xi,Zhe Jiang, Hu Bei,Shan Xi,Shan Dong,Yun Nan,Gan Su,Si Chuan,etc.In recent 20 years,it is well known from the medication in clinic in our affiliated hospital that Dung Beetle has a prominent effect in treating prostatitis and hyperplasia of prostate gland.Until now,there are less of researches in Dung Beetle's chemical components in domestic or abroad, which couldn't explain the application mechanisms in clinic and control the quality at the same time.To aim directly at these statuses,our experiences are based on the effect on anti-protatitis to screen the effective part with pharmacodynamics and study on the principal basis primarily in Dung Beetle.In the experiences,we adopted the Zhang Yaqiang's methods to make the rats contracted to chronic prostatitis,and screen the effective parts of anti-prostatitis.The results indicated that the lipid and water parts are effective.We had determined the amino acids in water part,where contains eighteen amino acids and among which there were seven essential amino acids in our body.We had determined the fatty acids with GC chromatography and isolated the chemical components with gel silica column chromatography in lipid part.From the results,we could know that there were nine fatty acids in lipid part, which contained unsaturated and saturated fatty acids.Otherwise,two compounds are finally isolated.The structures of these compounds were identified by means of NMR,MS,etc.The compounds were octadecanoic acid,cholesterin.They are isolated from Dung Beetle for the first time.Cholesterin as an index component to control the quality was detected by RP-HPLC,the average content of the cholesterin in five kinds Dung Beetle was 0.3381%.Finally,we drafted a quality specification protocol according to the experiences.In this study,we found the effective parts and the correlated components,which revealed the principal basis on anti-prostratitis on some degrees,established the scientific quality evaluation methods,and offered some scientific evidences and quality assurance in Dung Beetle's clinic application.
