Objective: Colorectal cancer is one of the most common malignant tumors, It is one of the main tumor which harm human health too, and it was the second tumor killer in western countries, the incidence rate was 50 per 100,000. In our country the incidence rate was 69.9 per 100,000(Shanghai Cancer Institute, 1999), It seized the fifth Position, With the development of people's life and culture degree, The dietary had changed greatly, People have more and more refined food and high-fat or high-protein food, Which resulted in the increasing of the colorectal cancer incidence rate. So we must find a new index to evaluate the early diagnosis, therapy and prognosis of colorectal cancer. Nuclear factor of kappa B was the key gene which regulated the genes transcription, which could be activated by many factors, and could control many genes expressions, for example Immunologic reaction, embryogenesis, apoptosis, tumor genesis, metastasis, physiologic reaction, pathologic reaction. NF-κB could control the excretion of intercellular adhesion molecule and extracellular matrix, which had the important effect in the metastasis of tumor. Matrix metaloproteinase was a huge family of proteolytic enzyme, According to the specificity of substrate, Which were divided for groups: collagenase, gelatinase, matrix degradation enzyme and MT-MMPs. Which could degradate many extracellular matrix Component, which have intimate relationships with the invasive and metastasis of human colorectal cancer, MMP-7 was the important one, It could degradate collagen IV, collagen V, laminin, fibronectin , gelatin in Interstitial space. MMP-7 have the close relationship with the tumor's invasive and metastasis. Nuclear factor of kappaB and MMP-7 play a important part in the occurrence, progression and diffusion of the neoplasm, The immunohistochemical SP was used to determine the NF-κB, MMP-7 expressions in the carcinomatous tissues of 50 patients and the tissues 3cm, 5cm, 10cm from the carcinoma edge. For the purpose of probing into the MMP-7 and NF-κB expressions in colorectal adenoca,and into the correlation between expressions of NF-κB, MMP-7 in the carcinomatous tissues of 50 patients and clinical pathologic factors of the cancer. Which could provided theorical basement for the diagnosis, therapy, prognosis of the tumor. Methods: Collect tissues from 50 colorectal adenoca patients by surgical operation. 39 male, 19 women. Age from 27-74 years old, the average is 53.5 years old. Among them 27 colonic adenocarcinoma, 23rectal adenocarcinoma, 18 Dukes classification A-B, 32 Dukes classification C-D, All tissue are divided into two degree, high and middle differentiation team(34),low differentiation team(16). None had accepted the treatment of any anti-tumor before operation. All of samples were fixed by neutrality formalin solutions, Embedded by paraffin, sliced in to 4μm slice,1 HE staining, 3 SP staining, examining Nuclear factor of kappaB and matrix metaloproteinase-7 expression in the tissue. Results: The positive product of NF-κB was mainly localized in the cytoplasm of tumor cells. And also in the nucleus, The positive product of MMP-7was mainly localized in the cytoplasm of tumor cells and in cell membrane. The positive color was Buffy. Scoring method: According to the percentage of positive cells, 0, positive cells <5%;1, positive cells 5%-24%;2,positive cells 25%-50%;3, positive cells >50%. According to the depth of Pigmenting, 1, weak Pigmenting; 2,middle Pigmenting; 3, deep Pigmenting .strong positive Pulsing the two fraction marks together, 0(-),negtive;<4(+),weak positive;≥4(++). NF-κB, MMP-7 expression rates were respectively 78%and86% in the carcinomatous tissues of 50 patients, respectively 68.0%,56.0%,23.5%and34.0%,28.0%,6.46% in the tissues 3cm,5cm,10cm from the carcinoma edge. The expression rate of NF-κB protein in colorectal adenoca was significantly higher than that in the tissue which was 10cm from the carcinoma edge(P<0.05).
