ObjectiveThe research was to investigate the effects of chinese herbal compound in rats with chronic renal failure(CRF) induced by adenine by observing rats' kidney function,the change of renal histomorphology,the change of anti-oxidation serum enzyme activity,expression of transforming growth factor-β1(TGF-β1),α-Smooth muscle actin(α-SMA) in the kidney,to explain the mechanism of chinese herbal compound on prevention and treatment of chronic renal failure,to provide experimental evidence for the further development and application of chinese herbal compound。Method48 SD male gender rats were divided into 6 groups:normal group,model group, niaoduqing group(0.2g/ml),chinese herbal compound high dosage group(5.04g/ml),chinese herbal compound middle dosage group(2.52g/ml),chinese herbal compound low dosage group(1.26g/ml),each group has 8 rats,each rat in model group,chinese herbal compound group,niaoduqing group was infused with adenine(200mg/kg/d) into stomach,in the same time,each rat in chinese herbal compound group was also infused with chinese herbal compound into stomach for 24 days,niaoduqing group was given niaoduqing liquid,the content of kidney function(serum creatinine,blood urea nitrogen),hemoglobin,red blood cell count,haematocrit,mean platelet volume, the change of anti-oxidation serum enzyme activity of superoxide dismutase(SOD) and changes in histopathology were observed after 24 days,expression of transforming growth factor-J31(TGF-β1),α-Smooth muscle actin(α-SMA) in the kidney were examined by immunohistochemistry。Results①Compared with the model group,renal weight,renal index,the level of Scr in each chinese herbal compound group,niaoduqing group were decreased significant (P<0.05);the level of BUN in the chinese herbal compound low dosage group,niaoduqing group were decreased significant(P<0.05);the level of serum SOD in the chinese herbal compound high,low dosage group,niaoduqing group were increased significant(P<0.05);expression of TGF-β1 in the chinese herbal compound middle, low dosage group,niaoduqing group were decreased significant(P<0.05);expression ofα-SMA in each chinese herbal compound group,niaoduqing group were decreased significant(P<0.05);the effect of low dosage group,niaoduqing group were best。②Compared with the model group,there is no apparent statistical significant difference in the level of HGB,RBC,HCT,MPV in each chinese herbal compound group,niaoduqing group(P>0.05)。③Pathological results of each chinese herbal compound group,the niaoduqing group were better than those of the model group, renal pathologic changes were palliated significant,the degree of injury in the low dosage group and niaoduqing group were the lowest。ConclusionChinese herbal compound can protect renal function of adenine-induced CRF rats,delay the development of CRF,depress the level of BUN and Scr,the mechanism may be related to suppressing the TGF-β1,α-SMA and antioxidation。...
