Yiqihuoxue Compound Study The Impact Of Chronic Heart Failure Rat Cardiac Ang Ii,

Purpose:To observe the associated impacts of benefiting Qi and activating Blood compound recipe to the myocardial angiotensinⅡ(AngⅡ) in chronic heart failure ,the subject establishes the chronic heart falling (CHF) model rats. From the gene and protein level, approaching the mechanism and target point of the Chinese medicine on CHF, and providing the scientific basis of preventing and curing the CHF.Material and method:60 Sprague SD rats were randomly divided into groups of Chinese medicine, Western medicine group, model control group and sham operation group 4. Application of coronary artery ligation with hunger with exhaustive style swimming and hunger methods successfully established the model of chronic heart failure. Chinese medicine group was fed with benefiting Qi and activating Blood compound recipe, western medicine group was fed with lisinopril, blank control group and model control group were fed with distilled water. All rats were killed after 6 weeks, whichever rat heart myocardium, detected by Western blot analysis of protein expression of AngⅡ, with real-time quantitative fluorescence PCR detection of AngⅡgene expression.Results:1.After the model of rats had made weight loss, depressed, coat color dim, and left ventricular ejection fraction calculation of EF values EF≤60% by Doppler echocardiography ,it signs the left heart failure2. Benefiting Qi and activating Blood compound recipe can make the AngⅡmRNA and AngⅡprotein expression decreased significantly, and stronger than the western medicine group. Conclusion:1. Application of coronary artery ligation with hunger with exhaustive style swimming and hunger methods can successfully establish the model of chronic heart failure. The model has advantages of reliable theoretical basis, convenient.2. Benefiting Qi and activating Blood compound recipe can inhibit the RAS system of chronic heart failure rats myocardium ,by reducing the rat myocardial tissue levels of AngⅡmRNA and protein expression of AngⅡ, which inhibit the RAS system, particularly the role of AngⅡon the heart, through the inhibition of the strong vasoconstriction, inhibition of myocardial cell proliferation, and other mechanisms to protect the cardiovascular damage and cure the heart failure. The effect of Benefiting Qi and activating Blood compound recipe is significant.
