Fructus corni is the dried and mature fruits of Cornus officinalis Sieb.et Zucc. which is a member of the family of Fructus corni Plant. Its action is to benefit liver and kidney and to put restraint on sperm and to solidify collapse. It has been used in China by traditional healers for the treatment of vertigo and tinnitus, aching pain in waist and knee, impotence and spermatorrhea, enuresis and urinary frequency, hematorrhea and leucorrhea, sweating and collapse, inter-heat and diabetes. In the recent years, it has been found by research that the extracted fractions from Fructus corni have therapeutical effects on hemorrhagic shock, immunoloregulation, inflammation, blood sugar, antioxidation, cancer, bacteria and etc.The main components of Fructus corni are known to contain iridoid glycisides, organic acids and volatile oils and etc. It is reported that loganin in Fructus corni is cardiovascular bioactive component.In the course of our research for antishock activity from Fructus corni, we found the extract solution from Fructus corni had significant antiarrhythmic effect. Thus, we separated different fractions from total extract solution in successive steps by using ether, ethyl acetate and n-butanol. The physical and chemical properties were investigated and the analytical methods and quality control were established.In the aspects of clinical pharmacology and mechanism of drug action, the pharmacokinetics of Chinese traditional medicines is showing its importance and necessity, especially for the study of pharmacokinetics on active components in Chinese traditional medicines, clarifying the processes of absorption, distribution, metabolism and elimination in thebody will have important significance in guiding rational use of medicines.PART ONE: STUDY ON EXTRACTION PROCESSES ANDQUALITY CONTROL OF THE EXTRACT SOLUTIONFROM FRUCTUS CORNIObjective: To study the optimal conditions and technology of preparation of the extract solution from Fructus corni and how to remove the impurity from the solution in order to obtain stable extract solution. To study the identification, examination and the determination of content for the extract solution from Fructus corni and to establish the standard of quality control in order to control the quality of the extract solution from Fructus corni effectively.Methods: The factors of the extraction including decoction time, decoction times and the amount of adding water were studied by using orthogonal experimental design according to the amount of the total glycosides determined. The optimal methods for removing protein, amylose and tannide were studied and the optimal pH and the amount of active carbon added and positive ion resin used were ascertained.The extract solution was further extracted in successive steps by using ether, ethyl acetate and n-butanol. Thus, the extract solution from ether, the extract solution from ethyl acetate, the extract solution from n-butanol and the residual solution were obtained respectively.Ursolic acid and loganin in the extract solution were identified by using Thin Layer Chromatography (TLC).The determination of content of the total organic acids in the extract solution from Fructus corni was established by using potentiometer titration and the precision and stability of the method were investigated.The determination of content of the total glycosides was studied by using spectrophotometry. The extract solution was diluted with water and10then the following step was to subject to 101 resin using a gradient of H2O and 70 per cent of alcohol to give total glycosides-containing fraction. The total glycosides were visualized by adding visualization reagent and then the solution was scanned and compare the maximum absorbance wavelength with that obtained from loganin, morroniside, panaxadial and geniposide and at last the detectable wavelength and the reference sample was ascertained. Under the condition, the factors including temperature and time to affect visualizing were studied.
