Objective:By investigating the expression of EZH2,Ki—67 in the normal gastric mucosa,juxtacancerous gastric mucosa and gastric adenocarcinoma,this study analyzed the difference of expression among two kinds of protein and the relation between the clinicopathologic characters of gastric adenocarcinoma with the protein's expression in order to explore the machinery of molecular biology for gastric adenocarcinoma.The experience would look for the new markers for evaluating biological characters and gene therapy of gastric adenocarcinoma.Methods:We collected 50 cases of gastric adenocarcinoma tissue,20 cases of juxtacancerous gastric mucosal tissue and 20 cases of normal gastric mucosal tissue. Then we examined the expression of EZH2,Ki—67 among the 90 cases of paraffin-embeded tissue.The results was treated by Wilcoxon test,Kruskal-Wallis test and Spearman Correlation analysis.Results:1.The expression level of EZH2 increased progressively from normal and juxtacancerous gastric mucosal tissue to gastric adenocarcinoma tissue.The positive immunostaining rate in the normal gastric mucosa,juxtacancerous gastric mucosa and gastric adenocarcinoma was 10.0%,45.0%,76.0%respectively,and the difference was significant among the three groups(P<0.01).The positive rate of EZH2 was lower in normal gastric mucosa than that in juxtacancerous gastric mucosa and gastric adenocarcinoma(P<0.05),and the positive rate of EZH2 was lower in juxtacancerous gastric mucosa than that in gastric adenocarcinoma(P<0.05).There was a relationship between the expression of EZH2 with the histological grade,lymph node metastasis,muscle invasive depth and clinical stage(P<0.05),but not with age,sex and position of tumor(P>0.05).2.The expression level of Ki-67 increased progressively from normal and juxtacancerous gastric mucosa tissue to gastric adenocarcinoma tissue.The positive immunostaining rate in the normal gastric mucosa,juxtacancerous gastric mucosa and gastric adenocarcinoma was 5.0%,40.0%,70.0%respectively,and the difference was significant among the three groups(P<0.01).The positive rate was lower in normal gastric mucosa than that in juxtacancerous gastric mucosa and gastric adenocarcinoma(P<0.05),The positive rate was lower in juxtacancerous gastric mucosa than that in gastric adenocarcinoma(P<0.05).There was a relationship between the expression of Ki-67 with the histological grade,lymph node metastasis,muscle invasive depth and clinical stage(P<0.05),but not with age,sex and position of tumor(P>0.05).3.The expression of EZH2 positively correlated with the expression of Ki—67 in gastric adenocarcinoma(r_s=0.695,P<0.01).Conclusion:1.The increasing expression of EZH2 could accelerate the cancerization of the normal gastric mucosal tissue.The high expression of EZH2 could accelerate the histological grade,clinical stage,lymph node metastasis,and muscles invasion depth of the gastric adenocarcinoma.2.The increasing expression of Ki—67 could promote the cancerization of the normal gastric mucosal tissue.The high expression of Ki—67 could be help to the histological grade,clinical stage,muscles invasion depth and lymph node metastasis of the gastric adenocarcinoma.3.The expression of EZH2 positively correlated with the expression of Ki—67 in the gastric adenocarcinoma tissue.This result approved that EZH2 make the function of promoting cancerization by regulating nagetively the expression of Ki—67 besides by its own action.
