Background:Gastric carcinoma is one of the most common malignancies of the digestive system,Seriously harmful to people’health.It’s the second leading cause of cancer mortality in the world.About one million people were diagnosed as gastric carcinoma[1]. The rate of the number in patients with gastric cancer in China for that in the world.The development of gastric carcinoma was correlated with the change of Gene regulation.Gastric carcinoma has high incidence and mortality rate and poor prognosis.Cyclooxygenase is one of the important inducible enzyme of Inflammatory process.It’s also a important rate-limiting enzyme of prostaglandin synthesis process,which can catalyze arachidonic acid conversion for gland products. So cyclooxygenase can participate many pathophysiological processes. Cyclooxygenase has two isozymes(COX-1and COX-2). Cyclooxygenase-1is structural enzyme,located in the endoplasmic reticulum and expressed in most tissue cells.Its key function was maintaining normal physiological function of cells and the stable internal environment. Cyclooxygenase-2is inducible enzyme,mainly located in the nuclear membrane.Product of prostaglandins could enter the nuclear and adjust the transcription of gene in target cells.many studies have shown thatcyclooxygenase-2overexpressed in tumor tissue and correlated with the development of tumor.Vascular endothelial growth factor is the strongest, the most specific and important regulatory factor.Recently,studies have shown that vascular endothelial growth factor is correlated with the department of tumor.but molecular mechanisms of VEGF in gastric carcinoma was not yet clear now. Study about The expression of COX-2and VEGF and their correlation in gastric adenocarcinoma is less at home and abroad currently.In this study we analyzed the relationship between Cox-2and VEGF and the clinicopathologic characteristics by using cases of large sample. That will provide some theoretical basis for learning more about the mechanism of the development of gastric carcinoma and assessing its biological behavior and prognosis in addition to carrying out the targeted therapyObjective:To investigate the expression and correlation of cyclooxygenase-2and vascular endothelial growth factor in gastric adenocarcinoma tissueMethods:Cyclooxygenase-2protein and vascular endothelial growth factor expression in478cases of gastric adenocarcinoma tissue and49cases of adjacent normal tissue were detected by using immunochemical staining and the relationship between Cox-2and VEGF was analyzed together with the clinicopathologic characteristics.Results:The positive rate of COX-2or VEGF in gastric cancer tissue were67.4%and63.2%.The expression of Cox-2and VEGF was associated with the grade of local infiltration,TNM stage,lymph node metastasis (P<0.05),while not with age,gender,type,histological grade,and the size of tumor(P>0.05).The expression of Cox-2was closely correlated with the expression of VEGF(r=0.329P<0.01). The positive rate of Cox-2and VEGF in gastric adenocarcinoma tissue was higher than which in adjacent normal tissue (P<0.01)Conclusion:The overexpression of Cox-2and VEGF associated with the grade of local infiltration,TNM stage,lymph node metastasis means that Detection of Cox-2andVEGF protein may be used to assess the malignant biological behavior and prognosis of gastric cancer.The significant relationship between overexpression of Cox-2and VEGF indicates that Cox-2and VEGF can regulate each other and induce the tumorgenesis and development together.
