Objective:The expression of EpCAM and MMP-9Protein in human gastricadenocarcinoma, dysplasia and normal gastric mucosa tissue was investigated to explorethe relationship of tumorigeesi, invasion and metastatic of gastric adenocarcinoma.Methods: Collected60cases of paraffin-embedded gastric adenocarcinomaspecimens and30cases of dysplasia gastric mucosa tissue and normal gastric mucosatissue of30cases of paraffin in our hospital pathology. Immunohistochemistry ofElivision method was used to detect the expression of EpCAM and MMP-9amongthem.Statistical softwere was used to analyse its expression and its correlation withclinicopathological parameters. The correlation between EpCAM and MMP-9were alsoanalyzed.Results:1. Positive rates of EpCAM in normal tissues, dysplasia tissues and gastricadenocarcinoma are6.7%、60%and86.44%,which shows gradual increasing trend.Difference between them was statistically significant (P <0.05). The expression ofEpCAM in gastric adenocarcinoma were relevanted to tumor cell diferentiation serosainvasion, tumor cell differentiation, lymph node metastasis and TNM staging. Thedifference was statistically significant (p<0.05). The expression of EpCAM wasuneorrelated with gender, age and tumor size(P>0.05).2. Expressions of MMP-9in normal tissues, dysplasia tissues and gastricadenocarcinoma are16.7%,33.3%and67.8%. The Expressions of MMP-9in gastriccarcinoma tissues was significantly higher than the normal tissues (P <0.05). It was closely correlated with tumor Serosa invasion, TNM staging and lymph nodemetastasis, the difference was statistically significance(P<0.05). The expression ofMMP-9was uneorrelated with gender, age, tumor size and tumor cell differentiation(P>0.05).3. Difference of EpCAM and MMP-9expression in gastric adenocarcinoma tissueswas a positive correlation between them (P<0.005,r=0.363).conclusion:1. Overexpression of EpCAM may be an early event of gastric adenocarcinoma.2. EpCAM and MMP-9may play an important role in invasion and metastasis ofgastric adenocarcinoma.3. There was a positive correlation between the expression of EpCAM and MMP-9ingastric adenocarcinoma significantly. The expression of EpCAM and MMP-9could beused to diagnose and predict the biological behavior of gastric adenocarcinoma.
