| Pomegranate is a kind of common fruit in our country.There are many studies about the effects of pomegranate in traditional Chinese medicine and other ethnomedicine,which focused on pomegranate juice pomegranate flower and bark.It was confirmed that pomegranate have many effects in its juice bark and flower by modern medicine practice such as disinfection,anti-inflammatory and helminthicide.In recent years,people extract pomegranate oil,which has been used as cosmetic and health food's materials.Since there are limited data about the biological efficacy of pomegranate,the consumers were confused by exaggerated propaganda.The present study is performed to explore the biologic efficacy of pomegranate oil in order to investigate pomegranate oil's biologic efficacy and provide experimental data for the further development and application.Objective:The present study is designed to analyze the fatty acid in pomegranate seed oil , investigate the biological efficacy of pomegranate such as anti-lipid peroxidation , decreasing blood fat and estrogenlike effect , and provide experimental evidence for its function study and experimental data for the further development and application.Methods:First of all,the fatty acid in pomegranate seed oil were detected.The pomegranate seed oil was put into KOH-Etoh solution to saponify and hydrolyze in order to extract fatty acids, then the fatty acids were methylated by H2SO4-MeOH(volume ratio 1:1)solution.The methylated fatty acids were analyzed by GC-MS.Second,the effect of pomegranate seed oil on lipid peroxidation was studied.Forty Wister rats (20-22 month) were divided randomly into 4 groups and fed with normal food.The control group rats were intragastric administration colza oil 2.0ml/kg bw at 8:00 am.Other groups were intragastric administration mixture of pomegranate seed oil and colza oil 2.0ml/kg by according to the ratio of 1:9,1:3 and 1:1.The rats were weighed once a week (empty stomach).After 4 weeks,these rats were killed and their blood and livers were isolated for further detection.The third experiment was designed to investigate the effect of pomegranate seed oil on blood fat of high fat diet rats.Fifty SD rats were divided randomly into 5 groups.The control group rats were fed with normal food,other groups were fed with high-fat food and were intragastric administration at 8:00 am.The high fat control group was intragastric administration with colza oil 2.0ml/kg;the other 3 groups were intragastric administration with mixture of pomegranate seed oil and colza oil 2.0ml/kg with the ratio of 0.5:1,1:1 and 1:0.5 respectively.The rats were weighed every 6 days.After 4 weeks,the rats were killed and their blood,fat and liver were isolated for further detection.Furthermore,the estrogenlike effect of pomegranate seed oil was studied with clinical cases.Total of 110 female patients,40-56 years old,who has either oophorectomize or climacteric symptom without organic disease were chose to divide into 2 groups randomly.The treatment group ate 4 pomegranate seed oil capsulae molles in 2 times every day.Placebo group ate placebo.They all stopped eating other health food,except of oophorectomize female didn't stop taking treated medicine.The index were determined after 5 weeks.Results:1.The average content of polyunsaturated fatty acid are about 90%,the content of punicic acid is 60-80%.2.There were significant increase of SOD and GSH in the blood serum of the rats who were treated with pomegranate seed oil,and the serum MDA decreased significantly.(P <0.01).The tendency of Liver homogenate's SOD, MDA and GSH is similar with blood serum's.3.In high fat control group,all of the TC,TG,LDL-C and AI were increased in blood serum,these indexes of pomegranate seed oil group are all lower than high fat control group's.4.The adipose layer of high fat control group obviously thicker than other groups.It demonstrates that using pomegranate seed oil can prevent the thickening of rats'adipose layer especially with high dose(P <0.05).5.The Kupperman score of treatment group decreased obviously(P <0.01),but the control group doesn't have significant change (P>0.05).Conclusions: 1.The composition of pomegranate seed oil is very complex, but most of them are punicic acid.2.The pomegranate seed oil has anti- lipid peroxidation effect to old rat.3.The pomegranate seed oil can decrease experimental animal's blood fat.4.The pomegranate seed oil can relieve women's climacteric symptom in clinical experiment. |
