The Empirical Study On The Effect Of Lonicerae And Forsythiae Powder In Different Decoction Time On Febrifacient Bandicoots' BT(Body Temperature) And CAMP(Cyclic Adenosine Monophosphate) In Hypothalamus And Adjusting Immunifaction To Rat

Objective: Lonicerae and Forsythiae Powder was created by Wu Tang.It is made up of flos, forsythia suspensa, platycodon, menthae, henon bamboo leaf, glycyrrhizae,schizonepetae,fermen- ted soybean and fructusan.《Mild illness strip distinguishing》named it cool and moderate prescription. Wu Tang made expatiation about decoction time and cpalendmeans."capiating when fragrance efferens,please do not boil too much"was writened after prescription.This method is the key point to get curative. Some people ignore the decoction clinically, which leads to get no effective.Naturally,they look down upon Lonicerae and Forsythiae Powder. Although another observe the decoction time,no one knows what boiling too much means.There is no final conclusion about what time is the best decoction time.We determin the best decoction time in long-term clinical practice,but not yet experimentize. The objective of this empirical study is to investigate the best decoction time, the effect of Lonicerae and Forsythiae Powder in different decoction time on febrifacient bandicoots'BT(body temperature) and cAMP(Cyclic Adenosine monophosphate) in hypothalamus, and the effectiveness of adjusting immunifaction of Lonicerae and Forsythiae Powder.Methods: (1) 96 qualified bandicoots were randomly divided into 12 groups, i.e.,normal control group,model group, aspirin control group, Lonicerae and Forsythiae Powder boiled after 3 minutes large dose,dominated dose,small dose group, Lonicerae and Forsythiae Powder boiled after 6 minutes large dose,dominated dose,small dose group, Lonicerae and Forsythiae Powder boiled after 12 minutes large dose,dominated dose,small dose group.There were 8 bandicoots in every group.Except for normal control, bandicoots got barm suspl 10ml/kg by hypodermic injection.Soon after bandicoots were measured BT per hour,keeping 7 hours. Bandicoots were fed with leechdom after 210 minutes and their hypothalamus were got to measure contents of cAMP.(2) 88 male rats qualified were randomly divided into 11 groups, i.e., normal control group,model group, Lonicerae and Forsythiae Powder boiled after 3 minutes large dose,dominated dose,small dose group, Lonicerae and Forsythiae Powder boiled after 6 minutes large dose,dominated dose,small dose group, Lonicerae and Forsythiae Powder boiled after 12 minutes large dose,dominated dose,small dose group. There were 8 rats in every group. Except for normal control,rats were posed immunity suppression model by cyclophosphamide. After lavaging Lonicerae and Forsythiae Powder to rats 7 days, the englobement clearance ability of rats'RES(reticuloendothelial system)to lazy carbo nodule in blood flow,the ability of rats'swimming endurance ,the quantity of T cell subpopulation in peripheral blood and the weight of immune organ of rats were determined. Results: 1.After Bandicoots fed with leechdom 30 minutes, Lonicerae and Forsythiae Powder didn't display the effect of cooling (p>0.05), after Bandicoots fed with leechdom 90 minutes, Lonicerae and Forsythiae Powder boiled after 3 and 6 minutes large dose,dominated dose display the effect of cooling(p<0.05), after Bandicoots fed with leechdom 150 and 210 minutes,all of Lonicerae and Forsythiae Powder groups display the effect of cooling (p<0.05).The effect of Lonicerae and Forsythiae Powder boiled after 3 and 6 minutes large dose is better. Except for Lonicerae and Forsythiae Powder boiled after 3 and 6 minutes small dose and 12 minutes large, dominated and small dose, Lonicerae and Forsythiae Powder groups could cut down the contents of cAMP in hypothalamu(sp<0.05).The effect of Lonicerae and Forsythiae Powder boiled after 3 and 6 minutes large dose is better.2.Lonicerae and Forsythiae Powder boiled after 3 minutes large dose, dominated dose, 6 minutes large dose can raise remarkably the swimming endurance ability of rats with immunity suppression(p<0.05).3. Except for Lonicerae and Forsythiae Powder boiled after 6 minutes small dose group, 12 minutes dominated dose,small dose group,the rest groups of Lonicerae and Forsythiae Powder can raise remarkably the englobement clearance ability of RES of rats with immunity suppression,the value of K andαwere raised up remarkably(p<0.05), which boiled after 3 minutes large dose and 6 minutes large dose were better.4. Lonicerae and Forsythiae Powder can raise remarkably the quantity of CD₄⁺ T cell in peripheral blood of rats(p<0.05),which boiled after 3 minutes large dose is better.Lonicerae and Forsythiae Powder has the tendency of raising CD8+ T cell in peripheral blood of rats,but the effectiveness is not remarkable.5. Lonicerae and Forsythiae Powder boiled after 3 minutes large dose can raise remarkably the weight of thymus and spleen(p<0.05)of rats.Conclusions: 1. Lonicerae and Forsythiae Powder has the effect of relieving fever, which boiled after 3 and 6 minutes is better.2. Lonicerae and Forsythiae Powder has the effect of cutting down the contents of cAMP in hypothalamus, which boiled after 3 and 6 minutes is better.3. Lonicerae and Forsythiae Powder has the effectiveness of raising immunifaction.Lonicerae and Forsythiae Powder can raise remarkably the swimming endurance ability of rats with immunity suppression and the effect of it boiled after 3 and 6 minutes is better; Lonicerae and Forsythiae Powder can raise remarkably the englobement clearance ability of RES of rats with immunity suppression and the effect of it boiled after 3 and 6 minutes is better; Lonicerae and Forsythiae Powder can raise remarkably the quantity of CD₄⁺ T cell and the specific value of CD₄⁺ T cell to CD₈⁺T cell in peripheral blood of rats and the effect of it boiled after 3 minutes is better; Lonicerae and Forsythiae Powder boiled after 3 minutes can raise remarkably the weight of thymus and spleen of rats.4.The effectiveness to CD₈⁺T cell of Lonicerae and Forsythiae Powder is not remarkably, which because of: (1)The number of rats is small.(2)Lonicerae and Forsythiae Powder rebound normal proportion of CD₄⁺ T/CD8+T by increasing CD₄⁺ T.5. To sum up,the effect of Lonicerae and Forsythiae Powder boiled after 3 minutes is best.
