Objection to research the therapeutical effect of glutathione on essential- hypertension and provide its mechanisms.Method sixty-three subjects with diagnosis of essential hypertension were selected in hospital patients, 31 male and 32 female. All these disease should be excepted: secondary hypertension,chronic heart failure,myocardial disease,diabetes,stroke,coronary heart disease,tumor,liver and renal inadequacy,severe other systemic disease, and have not received any antioxidants . the patients were divided into two groups: conventional treatment, aged from 50 to 75 years, mean age 66.64±5.78,smoking/non-moking was 9/19;Test group, aged from 51 to 75 years, mean age 65.31±6.24,smoking/non-moking was 12/23,and with glutathione 2400mg intravenous drip,once a day, for 14 days; Meanwhile thirty normal subjects were selected . aged from 49 to 75 years, mean age 64.43±6.78 ,smoking/non-smoking was 10/20 .the MDA,SOD,NO,brain natriuretic peptide levels in serum were measured, echocardiography( Ei,Ai,E/A,IVRT,DT)was examined pretreatment and posttreatment in each group. Record the data mentioned above and manage statistic analysis with SPSS 14.0.Result1,the levels of SOD,NO,MDA,BNP in serum are obviously different between essential hypertension and normal people(P<0.01).2,the levels of SOD,NO,MDA,BNP in serum changed in essential hypertension changed posttreatment (P<0.01),and the TG group has changed significantly from CG (P<0.01 ).3,Glutathione can improve ventricular diastolic function more obviously than conventional treatment.Conclusion the levels of SOD,NO in serum are obviously lower than normal people in essential hypertension, the levels of MDA in serum are higher than normal people(P<0.01), the patients with essential hypertension have high oxidative stress state; the levels of SOD,NO,MDA in serum changed in the two groups after posttreatment (P<0.01), but the TG has changed significantly from CG (P<0.01 ), the results indicated that glutathione can restore redox state, improve the function of endothelial cell, the mechanism is related to its anti-oxidation function; the level of BNP in essential hypertension is higher than the normal people, there are significantly decrease in the two groups after treatment, and the TG is more obviously, the mechanism maybe related to the improvement of the function of endothelial cell, the restitution of the high oxidative stress, the improvement of the left ventricular diastolic function and the control of the blood pressure.
