Objective To study the effects of Ferulic acid(FA)pretreatment on normal and ischemic/reperfusion injury isolated rat hearts.Methods Seventy-two SD rats were divided randomly into four groups :Control group(con);Ischemic/Reperfusion group (I/R); Ferulic acid group (FA1); Ferulic acid +ischemic/reperfusion group (FA2).Isolated rat hearts were perfused in the model of Langendoeff.The cardiac function of HR,LVSP,﹢dp/dtmax,t-dp/dtmax(ms) and -dp/dtmax were observed before and after administration to investgate the effect of ferulic acid.Results In FA1 group, LVSP,﹢dp/dtmax and -dp/dtmax was inhibited sigmifictly to compare with contral group(P〈0.05); Inhibition rate of LVSP showed the increase dependent on the dosage of ferulic acid, especially, in groups over the concentration of 10-5mmol/L had statistical significance(P〈0.05).The levels of +dp/dtmax and -dp/dtmax demostrated inhibition from the concentration of 10-6 mmol/L ,and got the peak of inhibition on the concentration of 10-4 mmol/L ,however,the action of inhibition descented on the concentration of 10-3 mmol/L and 10-2 mmol/L.Compared to contral group , the Inhibition ratio of﹢dp/dtmax and -dp/dtmax demonstrated statistical significance(P〈0.05)from the concentration of 10-5 mmol/L to 10-2mmol/L in FA1 group. In FA2 group , recovery rate of LVSP increased dependent on dosage from the concentration of 10-5mmol/L to 10-2mmol/L,and showed statistical significance from the concentration of 10-5 mmol/Lto 10-2mmol/L(P〈0.05)comparation with . I/R group .After 30min and 60min of reperfusion,﹢dp/dtmax of FA2 group had statistical significance from the concentration of 10-4mmol/L to 10-2mmol/L. About recovery rate HR, FA2 group was lower than I/R group after 30min and 60min of reperfusion,and demonstrated statistical significance(P〈0.05). Conclusion Ferulic acid can significently inhibit the function of normal isolated rat heart . Ferulic acid play a protective role from ischemic/reperfusion induced myocardiol damage in isolated rat heart.
