Objective: To study the expression of HIF-1αprotein and Glut-1 protein in breast carcinoma. Their relationships between the two factors and with clinicopathologic factors were analyzed. To investigate the effect of neoadjuvant chemotherapy (NACT) on the expression of HIF-1αand Glut-1 in breast carcinoma and determine whether there is a relationship between the two factors and the effect of NACT.Methods: Their expressions of HIF-1αand Glut-1 in 20 cases of tumor adjacent normal breast tissues, 60 patients with breast carcinoma and 48 patients who received NACT before and after the treament respetively were examined by immunohistochemical staining.Results: The positive expression rates of HIF-1αand Glut-1 in breast carcinoma was 68.3%,63.3% respectively, showing a significantly difference between breast carcinoma and adjacent normhal breast tissues(P<0.05). No significance was seen in the rate of expression with age, menstrual status, tumor size and PR (P>0.05) , the expression of HIF-1αwas significantly correlated with the lymphatic metastasis, surgical-pathologic staging, histological grade and ER (P<0.05). The expression of Glut-1 was not correlated with age, menstrual status, tumor size, PR and histological grade , but correlated with lymphatic metastasis, surgical-pathologic staging and ER. Expression of HIF-1αwas positively correlated with expression of Glut-1(r=0.461, P<0.001).The expressions of HIF-1αbefore and after NACT in breast carcinoma were 75.0% and 52.1% respectively, decreased significantly after NACT compared with before, especially in patients who react to NACT. But there is no such obvious change in other patients who react poorly to NACT. There is no obvious change in the expression of Glut-1 before and after NACT. Patients whose Glut-1 was positive had poorly react to NACT than that was negative (P<0.05)Conclusions: There are overexpresions of HIF-1αand Glut-1 in breast carcinoma, The expression of HIF-1αwas positive with expression of Glut-1, The variation of their expression may contribute to the pathogenesis, development and prognosis of the breast carcinoma. The expression of HIF-1αdecreased significiantly in ptients who react to NACT. The expression of Glut-1 may be considered as a new predictive indicator for the therapeutic effect of NACT.
