Objective To investigate the relationship between the distribution and quantity of dendritic cells(DCs)and memory T lymphocytes in the tumor site and the prognosis of gastric carcinoma.Methods Immunohistochemistry was used to count the S-100 protein positive DCs and CD45RO antigen positive T cells in 102 gastric carcinoma tissues.The experimental result was statistically analyzed.The relationship between immune cells in tumor site and prognosis of gastric carcinoma were studied.The specimens were divided into 3 groups according to the median of DCs and CD45RO~+T cells:group A with the DCs and CD45RO~+T cells both≥median,group B with the DCs≥median and CD45RO~+T cells<median,or the DCs<median and CD45RO~+T cells≥median,group C with both DCs and CD45RO~+T cells<median.Results Immunohistochemistry showed,the number of TIDC in high power fild was from 0.5 to 57.1,The median was 7.7/HPF in 102 gastric carcinoma tissues,and the number of CD45RO antigen positive T cells in 10 high power fild was from 0.36%/5HPF to 69.14%/5HPF,The median was 12.34 %/5HPF in 102 gastric carcinoma tissues.The number of DC and CD45RO antigen positive T cells had relationship with clinical and pathological features. Infiltration of DC and CD45RO antigen positive T cells could influence the prognosis and postoperative life span.The postoperative life span in DC high infiltrating group(the median of life span and five year survival rate was separately 77 months and 79.1%)was significantly delayed compared with DC low infiltrating group(35 months and 15.2%).Also the postoperative life span in CD45RO antigen positive T cells high infiltrating group(the median of life span and five year survival rate was separately 58 months and 65.3%)was significantly delayed compared with CD45RO antigen positive T cells low infiltrating group(39 months and 30.3%).In the carcinoma tissues the number of S-100 positive DCs and the number of CD45RO positive T cells was significantly correlated(r=0.446,P=0.000).A greater number of DCs and CD45RO antigen positive T cells simultaneously(A group:DC≥7.7个/HPF and CD45RO antigen positive T cells≥12.34%/10HPF) in the carcinoma tissues showed a longer postoperative survival time of the patients(the median of life span and five year survival rate was separately 77 months and 84.9%).However,a lower number of DCs and CD45RO antigen positive T cells simultaneously(C group:DC<7.7个/HPF and CD45RO antigen positive T cells<12.34%/5HPF)in the carcinoma tissues showed a shorter postoperative survival time of the patients(the median of life span and five year survival rate was separately 34 months and 14.3%).The survival curve of A group was significantly defferent from C group(P<0.001).Multivariate analysis showed that DC infiltration(P=0.000;OR,0.893;95%CI,0.850-0.939),and lymph node metastasis(P=0.006;OR,2.236;95%CI,1.257-3.976)were independent prognostic factors.Conclusion DC can be used as an independent prognostic factor.And combination of the S-100 protein positive DCs and CD45RO antigen positive memory T cells in gastric carcinoma tissues can be precise respone of immune state in gastric carcinoma tissues.The survival rate of the patients who had more intratumoral DCs and memory T cells is higher,which indicates that DCs and memory T cells play an important role in Anti-tumor immune response.The result provide experimential basement for selection of gastric carcinoma clinical treatment program.
