Objective:To explore the distribution of subsets of memory T cells (Tm) of gastric cancer patients in peripheral blood and tumor microenvironment, and to investigate the relationship between Tm cells and clinicopathologic factors of gastric cancer patients. Meanwhile, the significant alterations were observed in percentages of Tm in preoperative and postoperative peripheral blood. The correlationship between the subpopulations of Tm and dendritic cells was also been investigated in the peripheral blood and tumor microenvironment of gastric cancer.Methods:1. The peripheral blood samples of82cases of gastric cancer were collected from Tianjin cancer hospital. And70healthy volunteers enrolled as healthy controls. Flow cytometry was used to detect the expression of Tm and dendritic cells. The Tm subsets of gastric cancer patients during the periopeation were also measured by Flow cytometry.2. Then, we investigated the Tm and its subsets in the tumor microenvironment and analyzed the relationship between Tm subsets and clinicopathologic factors. The percentage of dendritic cell was also examined by flow cytometry. The association between dendritic cells and subsets of Tm in tumor micoenviroment were analyzed.Results:1. The percentages of CD4+Tm, CD8+Tm, CD4+Tcm and CD8+Tcm in peripheral blood from gastric cancer patients were statistically lower and the percentages of CD4+Tem and CD8+Tem were significantly higher than health controls.2. The proportion of CD4+Tcm and CD8+Tcm increased after radical tumor resection, while the percentage of the CD4+Tem and CD8+Tem decreased significantly. Both higher percentages of peripheral CD4+Tm and CD8+Tm associated with myeloid dendritic cells in peripheral blood.3. Tumor infiltrating CD8+Tm expressed both central and effector memory phenotypes, whereas CD4+Tm displayed predominantly an effector memory phenotype.4. Significant associations were detected between the peripheral CD4+Tm and CD8+Tm and clinical stage of gastric cancer patients, as well as the CD8+Tcm and clinical stage and nodal metastasis. Higher percentages of tumor infiltrating CD4+Tm and CD8+Tm were significantly associated with the early disease stage.5. Both higher percentages of CD4+Tm, CD8+Tm, CD4+Tcm and CD8+Tcm associated with myeloid dendritic cells in peripheral blood and tumor microenvironment.Conclusion:The proportion of Tm subsets may reflect the immune suppressive and immune response to the tumor associated antigen.Tm and subsets of Tm were good immune indicators for the disease stage of gastric cancer.
