The First Part EXPRESSION OF PSTAT-3 AND VEGF AND ITS RELATIONSHIP IN PSORIASIS ABSTRACTResearch Background Psoriasis is a skin disease under polygenic inheritance background with dysimmunity,characterized by hyperproliferation of the epidermis, angiogenesis and accumulation of inflammatory,whose pathogenetic mechanisms is still in nubibus.At present,investigation of pathogenetic mechanisms includes abnormal hyperplasia of epidermal keratinocyte,abnormal hyperplasia of microvessels and immunologic derangement.Abnormality of microvessels relations with genesis,development and recidivation of psoriasis,it plays an important role in invasion of psoriasis.Vascular endothelial growth factor(VEGF)is an important enhancing factor for angiogenesis,which can facilitate permeability of microvessels, disintegration and hyperplasy of vascular endothelial cell and induce angiogenesis,etc. Researchers think highly of relationship of VEGF and genesis and development proceeding of psoriasis.Signal transducer and activator of transcription is an important transcription factor in JAK/STAT signal pathway.Activated STAT-3 (Phospho-STAT-3)has a series of biologic activities:accommodate hyperplasia, apoptosis and differentiation of cell.Gene of VEGF is accommodated straightly by signal transducer and protein of STAT-3. Research objective The aim of study is to detect the expression of VEGF and PSTAT-3 in involved psoriasis vulgaris tissue,observe their level of expression between involved psoriasis vulgaris tissue and normal control skin and analyze their dependability.To investigate the role of PSTAT-3 and VEGF in pathogenesis of psoriasis.Research Methods The expression and allocation of PSTAT-3 and VEGF was determined with immunohistochemistry in samples of psoriasis vulgaris paraffin imbedding tissue from 25 patients with active stage psoriasis vulgaris and those of skin tissue from 10 healthy individuals.The expressed quantity of PSTAT-3 and VEGF was quantitated with image analysis technique,and the difference between involved psoriasis vulgaris tissue and normal control skin was analyzed.The expressed correlation of PSTAT-3 and VEGF in involved psoriasis vulgaris tissue was analyzed.Result VEGF was stained in the cytoplasm of keratinocyte in basal layer and superbasal layer in involved psoriasis vulgaris tissue,PSTAT-3 was stained in the nucleus of keratinocyte in prickle cell layer and granular layer.Expression of PSTAT-3 and VEGF in involved psoriasis vulgaris tissue was significant higher than that was in normal skin,whose disparity had statistical significance(P<0.01).The expression of PSTAT-3 was positively correlated with expression of VEGF in involved psoriasis vulgaris tissue(r=0.703,P<0.01).Conclusion①PSTAT-3 was highly expressed in nucleus of epidermal keratinocyte and it was not expressed in normal skin.②VEGF was highly expressed in the cytoplasm of pidermal keratinocyte and it was not almost expressed in normal skin.③The expression of PSTAT-3 was positively correlated with expression of VEGF in involved psoriasis vulgaris tissue.④PSTAT-3 might enhance hyperplasia of microvessels in involved psoriasis tissue by up-regulating the expression of VEGF and it played an important role in genesis and development of psoriasis. The Second Part THE EXPRESSION AND SIGNIFICANCE OF ANG-2 AND ITS RECEPTOR TIE-2 IN PSORIASISResearch Background Psoriasis is a common,chronic,inflammatory dermatosis with erythema and scaling papules,characterized by hyperproliferation of the epidermis,angiogenesis and accumulation of inflammatory.Microvessels of the papillary dermis in psoriatic lesions are elongated,tortuous,and dilated,which contributes significantly to the proinflammatory response.Angiopoietin(Ang)is recently recognized as a family of cytokine in secreted glycoprotein.Ang is tightly related to tumor angiogenesis and it is mainly expressed in tumor cells and the surface of vascular endothelial cell.Ang functions as ligands for tyrosinkinase with immunoglobulin-like and epidermal growth factor homology domains(Tie-2).Ang participate maturity of blood vessels,maintain integrity and stability of blood vessels, and it plays an important role in development of blood vessels,reconstruction of blood vessels,inflammation,cancer,etc.Especially Ang-2 is considered to be most important in the angiogenesis among Ang.Ang-2 and Tie-2 play an important role in the Angiogenesis,the remodeling of the vascular network and the repair of trauma.Research objective The aim of study is to detect the expression of Ang-2 and Tie-2 in involved psoriasis vulgaris tissue and to observe their level of expression between involved psoriasis vulgaris tissue and normal control skin.The role of Ang-2 and Tie-2 in genesis,development and angiogenesis of psoriasis was approached.Research Methods The expression and allocation of Ang-2 and Tie-2 was determined with immunohistochemistry in samples of psoriasis vulgaris paraffin imbedding tissue from 25 patients with active stage psoriasis vulgaris and those of skin tissue from 10 healthy individuals.Semiquantitative analysis was done by the intensity of dyeing and the amount of positive cells.And the difference between involved psoriasis vulgaris tissue and normal control skin was analyzed.Result The expression of Ang-2 was negative and Tie-2 was expressed by endothelial cells in the deep dermis in normal skin.Ang-2 was expressed by endothelial cells in the papillary dermis of involved psoriasis skin.Tie-2 was expressed by endothelial cells in the highly vascularized papillary dermis as well as by endothelial cells in other areas including the deep dermis.Expression of PSTAT3 and VEGF in involved psoriasis vulgaris tissue was significant higher than that was in normal skin,whose disparity had statistical significance(P<0.01).The expression of Ang-2 was positively correlated with expression of Tie-2 in involved psoriasis vulgaris tissue(r_s=0.73,P=0.005).Conclusion①Ang-2 was expressed by endothelial cells in the papillary dermis of involved psoriasis skin,and it was not expressed in normal skin.②Tie-2 was expressed by endothelial cells in the highly vascularized papillary dermis as well as by endothelial cells in other areas including the deep dermis.The expression of Tie-2 in involved psoriasis skin was significant higher than that was in normal skin.③Angiogenesis plays an important role in progression of psoriasis.The overexpression of Ang-2 and Tie-2 receptor is closely correlated with the angiogenesis of psoriasis.④Ang-2/Tie-2 receptor maybe a useful therapeutic targeted factor.
