Objective: To investigate the expression of Fas-associated protein with death domain Fas(FADD) and Caspase-3 in chorionic villi of normal early pregnancy and missed abortion in early gestation stage and their relationship with the levels ofβ-hCG in maternal's serum. Study on the correlation of FADD,Caspase-3 andβ-hCG with the pathogenesis of missed abortion.Methods: Expression of FADD and Caspase-3 in chorionic villi from 30 cases of normal early pregnancy (8-10 weeks pregnant) women (normal early pregnancy group) and 35 cases of missed abortion(missed abortion group) was detected by immunohistochemical technique.The levels ofβ-hCG in maternal's serum were detected by chemical immunofluorescence method.Results (1) There were positive expression of FADD and Caspase-3 in chorionic villi in normal early pregnancy group. FADD and Caspase-3 was located in syntrophoblast cells,cotytrophoblast cells. The expression of FADD and Caspase-3 was stronger in syntrophoblast cells.(2) There were no difference in FADD and Caspase-3 localization between two groups. The expression of FADD and Caspase-3 in chorionic villi in missed abortion group was significantly lower than that in normal early pregnancy group (P<0.01).(3) The level ofβ-hCG in normal early pregnancy women was 24525.46±20655.69mIU/ml. in missed abortion maternal's serum was 53708.31±43974.21mIU/ml, The level ofβ-HCG in maternal's serum in missed abortion group was significantly lower than that in normal early pregnancy group(P<0.01).(4) Relativity analysis of the level ofβ-hCG in maternal's serum and the expression of FADD and Caspase-3 in chorionic villi take on the negative relativity(P<0.01).Conclusion: (1) FADD and caspase-3 could play an important role in the apoptosis of trophoblastic cell. Fas-mediated pathway may be the primary apoptotic pathway in trophoblastic cell. (2) Apoptosis in the trophoblastic cell is probably one of the most important affections to hCG secretion. The increment of apoptosis may be detract the function in endocrine secretion of human placenta.The disequilibrium of secretion of the human placenta may play an important role in the the missed abortion.
