Study On The Association Between Missed Abortion And Apoptosis In The Trophoblastic Layer

Objective : To investigate the association between missed abortion and apoptosis in the trophoblastic layer by determining apoptosis and expression of proliferating cell nuclear antigen(PCNA), Survivin and P53 in placental trophoblasts of patients with missed abortion,and analyzing the interrelationship of the three .Method : Apoptosis and proliferation in early pregnancy chorionic villi, which were stained by Hematoxylin-Eosin, from 30 samples of surgical aspiration and 35 samples of missed abortion were random selected and studied. Histological changes were observed with light microscopy. Proliferating cell nuclear antigen(PCNA), Survivin and P53 were assayed by immunohistochemical staining.Result : In normal chorionic villi ,there was no pathological change in cell morphology,and there were small amounts of trophoblastic cells to occur apoptosis, principally in the syncytrophoblastic layer. But in chorionic villi after missed abortion , there were extensive pathological changes in cell morphology,and there were a devil of cytotrophoblasts and syncytrophoblasts to occur apoptosis, especially in the latter. In missed abortion, PCNA of cytotrophoblasts decreased in comparison with their counterparts in surgical aspiration (P < 0.01), and the one of syncytiotrophoblasts was slight . In both villi after missed abortion, Survivin was decreased while P53 was increased compared with their counterparts in surgical aspiration, which was in consist with apoptosis, there is statistical significance(P < 0.01 ) . Among the trophoblasts of 35 patients with missed abortion, Survivin have a negative correlation with P53 ,and have a positive correlation with PCNA;P53 have a negative correlation with PCNA .Conclusion : The increment of apoptosis may be an important pathologic process in missed abortion, and its mechanisms may have something to do with the abnormal expression and interaction of PCNA, Survivin and P53.
