| As a common malignant tumor in the world, gastric cancer(GC)is a malignant tumor derived from the epithelial tissue of the gastric mucosa, and has a high clinical incidence.According to authoritative reports, the incidence of gastric cancer ranks fourth among all malignancies, and the mortality rate ranks third.Among them, men are significantly higher than women in gastric cancer, and the incidence increases with age.Among gastric cancers, gastric adenocarcinoma is the most common malignant tumor. Most patients with gastric adenocarcinoma have no obvious early symptoms, and more patients have severe cancer cell invasion and metastasis at the time of consultation, which poses an extremely serious threat to human physical and mental health, and is the focus of cancer prevention and treatment in China. The abnormal proliferation of tumor cells and the dysregulation of apoptosis are important factors leading to the occurrence of malignant tumors. The tumor cells in the body are affected by various factors, resulting in their inability to apoptosis normally, which can further lead to the occurrence of tumors. Therefore, investigating the core molecules that regulate the apoptosis mechanism of gastric adenocarcinoma cells and their mechanism of action is of great importance for the early screening and diagnosis of gastric adenocarcinoma.Related studies have shown that tumor cell apoptosis is related to the regulation of multiple target genes.Among them, the death receptor pathway and mitochondrial pathway of apoptosis are the most popular research mechanisms in all current studies.FADD(Fas associated protein with death domain,Fas-associated death domain protein)is a new cytoplasmic connexin protein that exists in the cytoplasm and can interact with DD(death domain,Fas molecular death domain).DD in FADD and DD in Fas molecule activate Fas-Fas L signaling pathway in tumor cells in the process of interaction.Fas-Fas L signaling pathway belongs to death receptor pathway in apoptosis. There are few reports on the expression of malignant tumors in gastric adenocarcinoma in China, which are limited to protein levels.Caspase-7(cysteine protease-7 protein)is activated by the initiation factors Caspase-1 and Caspase-8.After activation,it performs apoptotic functions. It is located downstream of the apoptotic cascade and is the cell Key effector of apoptosis. However, there are few studies on the expression of Caspase-7 in gastric adenocarcinoma in China, and it is limited to the protein level.Regarding the correlation between the two expressions in gastric adenocarcinoma,no research report has been reported in this regard. In summary, this design mainly uses immunohistochemistry, technology, and technology to detect the expression of FADD at the protein and m RNA levels, and the expression of Caspase-7 at the protein and level in gastric adenocarcinoma tissue and normal gastric mucosa tissue adjacent to the cancer, respectively.A comprehensive analysis was performed on the correlation between the two,and the relationship between the two in gastric adenocarcinoma tissues and clinicopathological characteristics was studied. To explore the impact of FADD and Caspase-7 on the generation and development of gastric adenocarcinoma,so as to provide new theoretical basis for the diagnosis and treatment of gastric adenocarcinoma patients. Objective:The purpose of this study was to investigate the expression of FADD protein and m RNA and Caspase-7 protein and m RNA in gastric adenocarcinoma tissues and normal gastric mucosa tissues adjacent to the cancer. The relationship between clinicopathological characteristics and the two.Methods:1 The expression levels of FADD protein and Caspase-7 protein in 60 samples of gastric adenocarcinoma tissues and 50 samples of normal gastric mucosa tissues adjacent to the cancer were detected by immunohistochemical staining and Western blotting technology to investigate whether the expression of both in gastric adenocarcinoma tissues Have a correlation, and analyze the relationship between it and the clinicopathological characteristics of gastric adenocarcinoma.2 Using q RT-PCR technology to detect the expression levels of FADDm RNA and Caspase-7m RNA in 60 clinical gastric adenocarcinoma tissue samples and 50 normal gastric mucosa tissue samples adjacent to the cancer. Its relationship with the clinicopathological characteristics of gastric adenocarcinoma.Result:1 Through immunohistochemistry,it was found that the positive expression rates of Caspase-7 protein and FADD protein in gastric adenocarcinoma tissue were 46.7% and 38.3%,respectively.They were 68.0% and 60%,respectively.The positive expression rates of the two in gastric adenocarcinoma were down-regulated compared with normal gastric mucosal epithelial tissues without disease, and the statistical analysis was different(P <0.05).1.1 There is no relationship between the expression levels of FADD protein and Caspase-7 protein in gastric adenocarcinoma tissue samples and the age and gender of patients. There is no difference in statistical analysis. The analysis of data found that lymph node metastasis, degree of differentiation, clinical TNM stage, and depth of infiltration were all related to the expression levels of the two,and statistical analysis was different(P <0.05).1.2 There is a correlation between the expression of FADD protein and Caspase-7 protein in gastric adenocarcinoma tissue samples, and statistical analysis has differences(P <0.05).2 Through the detection and analysis of the results, the expression levels of FADD protein and Caspase-7protein in gastric adenocarcinoma tissue samples were lower than those in gastric adenocarcinoma tissues, and high in normal gastric mucosal epithelial tissues adjacent to the lesions, statistical analysis There are differences(P <0.05).2.1 The expression levels of FADD protein and Caspase-7 in gastric adenocarcinoma tissue samples have no relationship with the age and gender of patients.Statistical analysis has no difference, but the analysis data found that lymph node metastasis, differentiation, clinical TNM staging The depth of infiltration is related to the expression level of the two,and the statistical analysis is different(P <0.05).2.2 In gastric adenocarcinoma tissue samples, the expressions of FADD protein and Caspase-7 protein are correlated, and the statistical analysis is different(P <0.05).3 The results of q RT-PCR showed that the expression levels of FADDm RNA and Caspase-7 m RNA in gastric adenocarcinoma tissue samples were significantly lower than those of normal gastric mucosal epithelial tissues without disease, and the statistical analysis was different(P <0.05).3.1 The expression levels of FADD and Caspase-7 in gastric adenocarcinoma tissue have no relationship with the age and gender of patients.Statistical analysis has no difference, but the analysis data found that lymph node metastasis, differentiation, clinical TNM stage,The depth of infiltration is related to the expression level of the two,and the statistical analysis is different(P <0.05).3.2 The expression level of FADD m RNA in tissue samples of gastric adenocarcinoma patients is correlated with the expression of Caspase-7 m RNA,and the statistical analysis is different(P <0.05).Conclusion:1 FADD protein and m RNA are lowly expressed in gastric adenocarcinoma, and highly expressed in normal gastric mucosal epithelial tissue adjacent to the disease-free cancer, and are related to lymph node metastasis of gastric adenocarcinoma, clinical stage of TNM,degree of differentiation, and depth of invasion,so it is available and involved The occurrence and development of gastric adenocarcinoma.2 Protein and low expression in gastric adenocarcinoma, high expression in normal gastric mucosal epithelial tissue adjacent to the cancer, and it is related to lymph node metastasis of gastric adenocarcinoma, clinical stage of TNM,degree of differentiation, and depth of infiltration,so it is available and involved in gastric adenocarcinoma Occurrence and development process.3 At the protein level and gene level,it is suggested that there is a certain correlation between FADD and Caspase-7 expression in gastric adenocarcinoma tissue, and there may be a certain positive synergy. |
