| Objective:(1)To explore the expressions of TNF-αmRNA and IL-1βmRNA,IL-10mRNA in the rat adjuvant arthritis synovial,to approach their role in the disease process of rheumatoid arthritis(RA).(2)To explore the mechanism of conventional acupuncture and the Governor for Tongmai acupuncture treatment of adjuvant arthritis(AA)and the effect of comparison between the two,to approach the possible mechanism of acupuncture treatment of rheumatoid arthritis.Methods:Complete Freund's adjuvant immune Wister rats with adjuvant-induced arthritis,inflammatory arthritis rats were randomly divided into four groups:the control group,the model group,the conventional acupuncture group and the Governor for Tongmai acupuncture group.(1)To observe the joint swelling,Whitty multiple joint pain and arthritis index score;(2)To observe synovial pathological changes by light microscopy; (3)To observe the expression of cytokines TNF-αmRNA,IL-1βmRNA and IL-10mRNA in the synovial tissue by anti-retroviral-polymerase chain reaction(RT-PCR).Results:(1)Compared with the control group,model group joint swelling,whitty score and multiple joint pain and arthritis blank index increased significantly and had significant difference(p＜0.05,p＜0.01) The index of conventional acupuncture group and the Governor for Tongmai acupuncture group were lower than that in the model group and had significant difference(p＜0.05,p＜0.01);The difference between the Governor Tongmai acupuncture group and conventional acupuncture group was obvious(p＜0.05);(2)Model group,the conventional acupuncture group and for the transfer of the Governor Tongmai acupuncture group synovial organizations infiltration of inflammatory cells,vascular proliferation and pannus the formation of synovial hyperplasia;Acupuncture and for the transfer of conventional Governor Tongmai acupuncture treatment,the index decreased significantly compared with the model group were significantly different,for the Governor Tongmai acupuncture group compared with the conventional acupuncture group were significantly different;(3)Expression of TNF-αmRNA and IL-1βmRNA in model rats synovial membrane was higher than the other group,expression of IL-10 mRNA was lower than the other group,and the difference between modle group and blank group was obvious(p＜0.01);after treatment,expression of TNF-αmRNA and IL-1βmRNA in conventional acupuncture group and Governor Tongmai acupuncture group was lower than the other group,expression of IL-10 mRNA was higher than the other group and had significant difference (p＜0.05,p＜0.01);The difference between Governor Tongmai acupuncture group and conventional acupuncture group was obvious(p＜0.01)Conclusion:(1)Governor Tongmai acupuncture can improve the AA rats foot swelling of arthritis,multiple indices and arthritis joint pain flexor few;The Governor Tongmai acupuncture method is better than conventional acupuncture method.(2)Conventional acupuncture Governor Tongmai acupuncture is better than conventional acupuncture in reducing rat synovial organizations infiltration of inflammatory cells,vascular proliferation and pannus formation.(3)Governor Tongmai acupuncture is better than conventional acupuncture in reducing the expression of synovial tissue TNF-αmRNA,IL-1βmRNA and increasing the expression of IL-10 mRNA. This maybe one of the mechanisms of acupuncture treatment of RA at the molecular level.(4)It has very clearly adjustment effect ofthe lever and the immunity disharmony of AA by the acupuncture,to cure the RA and reach to natural state of the Yin and Yang Equilibrium by the Strengthening Body Resistance to Eliminate Pathogen in the traditional Chinese medicine mechanism.
