Background and purpose:Intracerebral hemorrhage(ICH)is a common and often fatal stroke subtype,and an increasing number of people are paying close attention to it because of its high morbility, mortality and mutilation rate.However,currently there is no medical therapy available for these patients except blood pressure reduction and neurosurgical evacuation of the hematoma.After the classic prescription-Buyang Huanwu Decoction invented by Wang Qingren,a famous doctor of Qing dynasty,who initially posed the view of Qi-tonifying and stasis-eliminating therapy.Which has been widely used in clinical.Recently,the exprimental researches of Buyang Huanwu Decoction were focused on reducing cerebral edema,opposing oxygen-derived free radicals and apoptosis and inhibiting inflammatory reaction.Angiopoietin-1,an endothelium-specific proangiogenic factor, does play a crucial role in mediating reciprocal interactions between the endothelium and surrounding matrix and causing maturation and stabilization of vessels by virtue of phosphorylating its endothelial-specific receptor tyrosine kinase Tie-2.Recent studies have proved that in the central nervous system Ang-1 and Tie-2 were observed to be involved in a wide variety of disorders,ranging from brain tumors to cerebral ischemia.However,so far there has been few report about the expression of Ang-1/Tie-2 following ICH.Accordingly, the purpose of our present work is to evaluate whether the expression of Ang-1 and its receptor Tie-2 is altered in rat brains with collagenase-induced ICH,and tried to investigate effects of the classical prescription of Qi-tonifying and stasis-eliminating therapy, Buyanghuanwu decoction(补阳还五汤),and its Qi-tonifying(QT) component,stasis-eliminating(SE)component on expression of Ang-1/Tie-2mRNA following ICH,to elucidate its mechanisms in reconstruction of microvascular networks,to provide some new theoretical proofs for the clinic application.Methods:ICH model was induced by injecting collagenase typeⅦstereotaxically into fight globus pallidus.One hundred and fifty Sprague-Dawley rats were randomly divided into five groups,including sham control group(n=30),ICH model group(n=30),QTSE-treated group(n=30),Qi-tonifying(QT)-treated group(n-30)and stasis-eliminating (SE)-treated group(n-30),respectively administered orally with the classic QTSE prescription,Buyang Huanwu decoction,its Qi-tonifying and Stasis-eliminating components,all at the dosage three times that of a 70kg adult;While animals of sham group and ICH model group were administered orally with equal distilled water.The positional variations of the expression of Ang-1 and Tie-2 mRNA were assayed by reverse transcription-polymerase chain reaction(RT-PCR)at dayl,4,7,14,21 and 28d after the onset.Results:There were no significant expressions of Ang-1 and Tie-2mRNA in rat brains of sham group during the experiment. However,the expression of Ang-1 and Tie-2 mRNA in ICH model group and QT -treated group was observed in rat brains at 1 day after ICH.Furthermore,the expression increased gradually from day 7,and persisted at day 28(P<0.05).In QTSE -treated group,the expression of Ang-1 and Tie-2 mRNA was significantly higher than other three groups from day 7(P<0.05).Conclusions:ICH could lead to the upregulation of the expression of Ang-1 and Tie-2mRNA in intracerebral hemorrhagic rats that could be promoted by Qi-tonifying and stasis-eliminating therapy,which may be related with its promoting neurological recovery after the stroke.
