Background and purpose:Intracerebral hemorrhage (ICH) is a common and often fatal stroke subtype, and an increasing number of people are paying close attention to it because of its high morbility, mortality and mutilation rate. However, currently there is no medical therapy available for these patients except blood pressure reduction and neurosurgical evacuation of the hematoma. ECM (Extra cellular matrix ECM) mainly contains collagen (collagens), connected glycoprotein (including Laminin, Fibronectin) and other structural components of a mixture of insoluble tissue-specific three-dimensional structure,which is interaction with angiogenesis in the process of microvascular reconstruction after intracerebral hemorrhage.Angiogenesis can lead to the degradation of ECM components, remodeling, and ECM is important on cell proliferation, migration, survival, and forming new blood vessels and maturation.After the classic prescription-Buyang Huanwu Decoction invented by Wang Qingren,a famous doctor of Qing dynasty, who initially posed the view of Qi-tonifying and stasis-eliminating therapy.Which has been widely used in clinical.Methods:ICH model was induced by injecting collagenase type VII stereotaxically into right globus pallidus.One hundred and fifty Sprague-Dawley rats were randomly divided into five groups, including sham control group(n=30), ICH model group(n=30), QTSE-treated group(n=30),Qi-tonifying(QT)-treated group(n=30)and stasis-eliminating (SE)-treated group (n=30), respectively administered orally with the classic QTSE prescription, Buyang Huanwu decoction, its Qi-tonifying and Stasis-eliminating components, all at the dosage three times that of a70kg adult; While animals of sham group and ICH model group were administered orally with equal distilled water. The expressions of C-IV、 FN、LN-1and LN-5were evaluated by immunohistochemical method, and the number of positive expression in angiogenesis around the hemotoma at3rd、7st、14st、21st、28st days were analyzed semi-quantitatively.Results:There were no significant expressions of extracellular matrix components in rat brains of sham group during the experiment. However, the expression of C-IV in ICH model group was observed in rat brains at3day after ICH, reached to peak at day21, dropped down from day28. In QTSE-treated group, the peak expression of C-IV persisted to day21and higher than model group on many time points; the expression of FN in ICH model group was observed in rat brains at3day after ICH, the expression increased gradually from day7, and persisted at day21, dropped down from day28. In QTSE-treated group, the expression of FN is higher than model group on many time points. The expression of LN-1in ICH model group was observed in rat brains at3day after ICH, increased at day7, achieved peak at day21, dropped down from day28. In QTSE-treated group, the expression of LN-lwas higher than model group on many time points. The expression of LN-5in ICH model group was observed in rat brains at3day after ICH, reached to peak at day21, dropped down at day28,In QTSE-treated group, the expression of LN-5was higher than model group on many time points.Conclusion:1.The expression of the extracellular matrix components C-IV、FN、LN-1and LN-5were promoted around the hematoma after intracerebral hemorrhage. Probably because they are participate in angiogenesis of pathological tissues after intracerebral hemorrhage.2.The qi-tonifying and stasis-eliminating therapy can significantly can promote the expression of C-IV、FN、LN-1and LN-5, also improve the angiogenesis in the hematoma after intracerebral hemorrhage, which may accelerate the angiogenesis in the reconstruction of microvascular network in the damaged zone, and facilitate the repair of damaged tissue.
