Presently hypoxic-ischemic encephalopathy(HIE)is the most common fatal cause of infant death and morbidity.Traditional Chinese medicine consider hypoxia causes poor movement of blood and qi hence produces blood stasis and furthermore qi stasis.Currently on basis of Western medicine treatment,promoting blood circulation and removing blood treatment of traditional Chinese medicne is combined. Treat medium to severe HIE with Integrative Medicine is the developing orientation, countrymen's demand for traditional medicine promotes this process.It has been found that promoting blood circulation and removing blood treatment can improve brain microcirculation,preventing neurocyte from apoptosis and necrosis.However, there is still no further study about healing effect of invigorating qi and enriching the blood treatment on neurocyte.Modern neuropathology has confirmed that acute hypoxia induces accumulation of lactic acid,failure of natrium-kalium pumps, denaturalization and necrosis of cells,release of a large amount of cytokines,all these factors interact to accelerate rehabilitation of neural tissue.Tissues from different groups are tested for the amount of nerve growth factor(NGF),a representative factor of those cytokines,and try to unveil the relationship between endogenic cytokines and invigorating qi and enriching the blood treatment.Hopefully reveal the mechanism about how Chinese medicine improves neural tissue rehabilitation.By adding invigorating qi treatment of traditional Chinese medicne to understand its medium and long term neural function healing effect and instruct clinical use of Chinese medicine.Materials and methodsExperiment animal:80 healthy clean SD rats,off weaning for two weeks, weight about 100g,half male half female.Divided into 5 groups,each 16,8 among each are executed in every time point.Bilateral carotis communis ligature are performed in surgical groups to establish HIE model.After surgery,respectively stomach lavaged with normal saline(model group),xuefuzhuyu decoction(Chinese medicine groupl),xuefuzhuyu decoction plus 10g radix astragali(Chinese medicine group2)and xuefuzhuyu decoction plus 40g radix astragali(Chinese medicine group3).Observe neurological behaviors of those groups on the 1st day and the 6th day,apply Zea Longa neural function grading method.Obtain the brains after dislocation,observe pathologic changes in brain tissues after HE dyeing under light lens and test the amounts of NGF mRNA with quantitative fluorescence RT-PCR.In sham surgery groups the same procedure are performed,only left both sides of the carotis communis with no ligature.After surgery,respectively stomach lavaged with normal saline(control group).Observe neurological behaviors of those groups on the 1st day and the 6th day,apply Zea Longa neural function grading method. After dislocation obtain the brains,observe pathologic changes in brain tissue after HE dyeing under light lens and test the amounts of NGF mRNA with quantitative fluorescence RT-PCR.Data processing:Analyze with SPSS10.0 statistic software pack,datas are showed as mean values±standard deviation(x±s),mono-factor analysis of variance(ANOVA)between groups,while t test between paired samples.Datas were tested by Kruskal-Wallis test between groups,Mann-Whitney test between paired samples.All those tests take P<0.05 as statistically meaningful.Results1.Neurological behavior change:On the 1st day after surgery,rats from surgical groups represent various degree of neurological malfunction,no significant difference between Chinese medicine treated groups and modle group,while significant difference has shown between surgical groups and sham surgical group(P<0.05).On the 6th day,there is significant difference between Chinese medicine treated groups and modle group(P<0.05)while no significant difference between Chinese medicine groups.Contrast the 1st day,no remarkably improvement has been found in model group,remarkably improvements show in Chinese medicine treated groups(P<0.05) and there is no significant difference between groups on the 6th day.Neural function of sham surgery groups are normal with 0 score.2.Pathologic changes of brain tissues under light lens after HE dyeing:rats from surgical groups have the similar pathologic appearance,typical damages of HIE are witnessed:hyperplasia of cortex small neuroglia cells;shrinkage,vacuolation of hippocampi neurons,decrease or disappearance of Nissl bodies,karyodieresis;brain interstitial edema;narrowing of blood vessel cavicity,enlargement of the interstitial around gore;significant vacuolation of nerve fiber within infarction area.On 6th day past sugery,no abservable pathological improvement show in brain tissue from model group;on the other hand Chinese medicine groups improved remarkably,mainly rehabilitating changes of neurons.Sham surgery groups exhibit normal neural morphology,no visible pathological damage.3.Test the amounts of NGF mRNA with quantitative fluorescence RT-PCR.,results are shown as the differences between NGF andβ-actin PCR in pairs.Statistic displays that there is no significant difference between different Chinese medicine group 1 and model group,significant difference between another Chinese medicine groups and model group on 1 st day past surgery(P<0.05)while significant difference has shown in contrast to sham surgical group;on 6th day past surgery there is significant difference between all Chinese medicine groups and modle group(P<0.05) and there is significant difference from sham surgery group(P<0.05),no significant difference between Chinese medicine groups,no significant difference between model group and sham surgery group.There is significant difference between the 1st day and the 6th day past surgery in model group and there is no significant difference in other groups.Conclusion1.Stomach lavaging xefuzhuyu decoction can considerably improve neurological behavior changes and pathological changes in rats suffered HIE brain damage.2.Stomach lavaging of xuefuzhuyu decoction without radix astragali or with 10g radix astragali or with 40g radix astragali can strongly enhance NGF expression in rats brain tissue post HIE.3.There is no significant different ability of enhancing NGF expression in rats brain tissue post HIE among in stomach lavaging of xuefuzhuyu decoction without radix astragali and with 10g radix astragali and with 40g radix astragali.
