Objective To investigation on hisopathology changes in neonatal mice by hypoxic-ischemic. Methods the normal 7-day-old Wistar mice were studied. After unilateral common carotid artery was ligated and cut, the mouse was put in the hypoxia- box of 37.0℃and 8% hypoxia,92% nitrogen for 120 minute.The brain was collected at 7day,14day,30day after injury. The neuropathology were observed(HE,immunohistochemistry,electron microscope).Results (1)Hematoxylin and eosin:The trial group on 7day unilateral carotid ligation side displayed neurocytes arranged disorder, some neurocytes degenerated,cells swellsd,inflammatory cell infiltrated;On 14day displayed small capsular space formed, interstitial tissue was edema clearly;On 30day displayed many capsular space formed. At different time contrast with the trial group,the contral group showed the neurocytes eumorphism,no interstitial tissue edema and capsular space formed.(2)Immunohistochemistry:①MBP staining:The trial group MBP positive neurocytes in the periventricular zone on 7day were 3.07±2.08,on 14day were 3.07±1.8,on 30day were 4.03±1.94,to compared with the control group on 7day were 6.17±2.49,on 14day were 12.40±3.45,on 30day were 15.60±5.14 ,were decreased, P<0.05;②GFAP staining:The trial group GFAP positive neurocytes in the periventricular zone on 7day were 7.30±3.54,on 14day were 14.03±3.16,on 30day were 22.43±3.60,to compared with the control group on 7day were 2.33±1.73,on 14day were 2.73±2.21,on 30day were 3.37±2.16 ,were increased,P<0.05.(3)Electron microscope:The trial group on 7day neurocyte was edema, part of mitochondria was vacuolus, caryotin was condensed; 30day the neurocyte was irregulated obviously,part of nuclear membrane break down with content leakaged;The control group cytomembranes were integrated, kytoplasm was abundant, mitochondria and endoplasmic reticulum was normal,caryotin homogeneous distribution,nuclear membrane was integrated.Conclusion After hypoxic - ischemic in neonatal mice,the hisopathology changes on the unilateral carotid ligation side displayed periventricular cells swellsd,degenerated and leukomalacia formed.
