Objective:To explore Mahuangtang(Ephedrae Decoction)' s anti type I allergy mechanism through an experimental study of its effect on labrocyte degranulation in vitro,and to provide evidence for clinic usage of Mahuangtang in the treatment of allergic rhinitis.Method:Prepared the decoctum of Mahuangtang according to Treatise on Febrile Diseases(the proportion of ephedrae,ramulus cinnamomi,apricot seed,prepared radix glycyrrhizae was 9,6,6,3,respectively),and condensed the crude drug to the concentration of 50mg/ml in vacuum packaging.Resuscitated and cultivated labrocytes(RBL-2H3 cells)to their second generation living cells to 1×106/ml in concentration. Collected the second generation living cells and adjusted the concentration to 5×105 cells/ml,divided them into five groups.Among them,three groups were added to Mahuangtang with the concentration of 10-1mg/ml,10-3mg/ml,10-5mg/ml,respectively,another group was added to 10-3mg/ml Dexamethasone as a positive control,the rest one group was a blank control as nothing added.Added C48/80(10μg/mi)to induce the RBL-2H3 cells degranulation after 60 minutes cultivation,added Annexin V to the suspension and mix it slightly,standing for 10 minutes at ordinary temperature.Detected the percentages of labrocyte degranulation of each group by the flow cytometer within 30 minutes. Repeated the trial 6 times and got the average percentages(mean±SD)of the 5 groups.The data were analyzed by Concise Statistics 2000.When equal variances assumed,t test was conducted.When equal variances not assumed, t' test was conducted.P<0.05 was considered statistically significant.Result:The percentage of labrocyte degranulation of high concentration Mahuangtang group was 5.5%±0.40%,that of medium concentration group was 9.8%±0.57%,and low concentration group,17.6%±0.85%,the percentage of the positive control group was 1.8%±0.09%,that of the blank control group was 27.0%±1.60%.There were statistically significant differences within the 3 different concentration experimental groups(P<0.001).Differences was statistically significant in the high concentration Mahuangtang group and the positive control group (P<0.05),and in the low concentration Mahuangtang group and the blank control group(P<0.001).Conclusion:Mahuangtang had an inhibition effect on labrocyte degranulation to some extent,the higher concentration,the stronger effect.The high concentration Mahuangtang group(10-1mg/ml)had the strongest effect on inhibiting labrocyte degranulation,but still weaker than the Dexamethasone group.
