Objectives: To investigate whether the transcription and expresson of platelet-derivered endothelial cell growth factor in carcinoma of oesophagus cancer cell could be inhibited by RNA interference ,to observe whether silence the PDECGF gene specificly through in vitro exprement.Methods: There were 3 groups in our study,control group, vector group and siRNA group. desigening and synthesizing the pPDECGF shRNA plasmid to transfect human oesophagus cancer cell line,setting up stable expression after screen positive colone.After the transfection,the cell proliferation,invasive siRNA group and protein expressioning level were observed by MTT,RT-PCR and western blot.Results: In the siRNA group,the OD-value were much lower than the other two groups(P<0.05). Invasive ability were lower in siRNA group than in the other groups (P<0.05). PDECGF protein expressing level was lowest in the siRNA group. (1) the expression of PDECGF gene in oesophagus cancer was suppressed by Anti-PDECGF gene RNA interference vector pPDECGFRNAi, The PDECGFmRNA/β-actin decreased significantly in oesophagus cancer cells in siRNA group compared to other two groups,the difference shows significant stastitics(P<0.05).the difference of idler vetor group, and modle group shows no significant stastitics (p>0.05); (2) siRNA can suppress the proliferation of oesophagus cancer cell in vitro,the cytoactive , soft agar colon formation rate and cell migration rate decreased significantly compared to other two groups.Conclusion: The results suggest that siRNA of PDECGF genes can effecialy downregulate the expression of PDECGF genes and inhibite the proliferating and invasiving ability of oesophagus cancer cells.
