PARTⅠTHE DETECTION OF IP-9 AND IP-10 STIMULATED WITH IFN-γIN THE PRIMARY RAT HEPATOCYTE IN VITROObjective: To detect the mRNA level and the protein level of IP-9 and IP-10 stimulated with different concentrations of IFN-γin the primary rat hepatocyte in vitro.Methods: By the method of semi-in-situ collagenase perfusion, the primary rat hepatocyte was isolated and cultured. After the stimulation with the different concentrations IFN-γ(1ng/ml,10ng/ml and 100ng/ml ),the supernatant and cells were collected at different time points of 1h,12h,24h,48h and 72h, respectively. The PCR primers of IP-9 and IP-10 were designed and synthesized. The mRNA levels of IP-9 and IP-10 were detected via RT-PCR; the protein levels of the both chemokines were detected via ELISA.
