objective:the role of the PDECGF including, raise the synthesis of DNA, increase the ability of migration of the endothelia cells, stimulate the angiogenesis and tumor growth .the publicated literature show the cell that transfected the corresponding plussense and antisense RNA to mRNA can degrade the mRNA specifically. morever silence the corresponding gene. such post-transcriptional gene silencing was known as the RNA interference. in the recent year, RNAi technology has been paid more attention in mammalian cell and become the new important method on the gene function. to investigate the effect of the PDECGF on the tumor ,differentiation, migration of the bladder cancer, we adopt the gene silence method---RNA intervene suppress the PDECGF expression of the bladder cancer cell T24.according to the gene order of the PDECGF, construct the vector plasmid that can form the hairpin structure with the transcription vector pRNAT-U6/Neo containing the RNA polymerase promoter III, transfect it to bladder cancer cell T24 and observe the effect on the expression of the PDECGF. to observe if it can silence the PDECGF gene specifically though the in vitro experiment and bearing cancer naked mouse experiment in vivo as well as investigate the effect on the bioactivity of bladder cancer cell and the suppress extent on the tumor in vivo, morever establish the basis of progress and development of bladder cancer.method: the expression of PDECGF in bladder cancer and innocent bladder tissue were detected by immunochemistry method, compare the expression...
