Objective To study the regulation of shive-stopping capsule on expression of monoamine oxidase B (MAO-B) and tyrosine Hydroxylase(TH ) in rat brain .Methods The Parkinsonism model was prapaered by injection of 1% 6-OHDA in rat brain for two weeks. The tests were completed in 3 deffrent groups(Parkinsonism model group, normal controlled group and shive-stopping capsule treated group). The gene expression levels of MAO-B and TH in experemental rat brain were determined using RT-PCR. Moreover, the effect of shive-stopping capsule on activity of MAO-B in rat brain was determined by biochemical method.Results In comparing to PD model group, it showed that shive-stopping capsule could obviously inhibit the increase of MAO-B gene expression level in rat brain. It was 4.728times lower than that in PD model group. Furthermore, it showed that the effects of shive-stopping capsule on activity of MAO-B is in accordance with mRNA level detected by RT-PCR, and the inhibitary rate of shive-stopping capsule on activity of MAO-B was over 40 percent. On the contrary,the gene level of TH was obviously increased in rat brain which were treated with shive-stopping capsule. In comparing to PD model group, it was 2.195times higher than that in PD model group.Conclusion Shive-stopping capsule played the positive regulatory roles at the level of TH mRNA expressions in PD brain, and a negotive roles at the gene expression and activity of MAO-B. So it was suggested that shive-stopping capsule can be used to treat the PD in future.
