The pathogenesis of Parkinson's disease which is not yet difined is very complex. Popular theories include oxidative stress, apoptosis, gene mutation, immune abnormalities. Currently, Parkinson's disease can not be thoroughly cured, but only be controlled and relieved by western medicine, surgery, and other methods. In recent years, civil scholars began to study the treatment of traditional Chinese medicine on Parkinson's disease, and have made some progress. This paper studies the influence of tremor-stopping capsule on the signaling protein gene expression in substantia nigra part of 6-OHDA-induced PD model rats'brain from the perspective of Chinese medicine.Objective:discuse the influence of tremor-stopping capsule on the Several Cell Signaling protein gene expression in PD rats'brain, and further elucidate the pathogenesis of PD.Method: To choose 10 clean pure SD rats as the normal control group at random whose weights varie from 150 to 160g from 70 rats,the other 60 rats are used to prepare PD model.and inject 6-OHDA(6-Hydroxy dopamine) in the Substantia nigra part of the rats'brain to induce the symptoms of Parkinson's disease by using three-dimensional locational method. treat rats that are compliant to standards to 0 day, 7 days, or 14 days, remove their brain in a sterilized way after killing them through dislocation of cervical vertebra and extract total RNA from the cerebrum tissue, and furtherly transcript the contained mRNA into cDNA inversely, amplify genes in each group by using Reverse transcription polymerase chain reaction technique(RT-PCR)for gene amplification(TNF-RΙ,Fas,p53,Casepase-8 and Cytochrome c). analyse the results of PCR amplification by agarose gel electrophoresis, and detect each group's average optical density of amplified genes through Gray-scale analysis. analyse the differences of each group in the aspect of gene expression through statistical method, so that the influence of tremor-stopping capsule on the signaling protein gene expression in substantia nigra part of rats'brain can be known.Result: As can be seen from Agarose gel electrophoresis and statistical analysis, the objective genes expression of model groups increases magnificantly when compared with normal ones(P<0.01). The level of objective genes in model groups decreases after 7 days'treatment, and there is difference in Cytochrome c(P<0.05). Obvious differences can be seen among other four genes(P<0.01). They all have stastical magnification. When furtherly treated to 14 days, the expression of objective genes continue to decrease. The difference which is apparent has statistical magnificance(P<0.01).Conclusion:①6-OHDA can induce the activation of cell membrane receptor TNF-RΙand Fas, activate dead signal, and thereby cause the increasing of cell apoptosis regulated gene expression, and lead to apoptosis of cell.②tremor-stopping capsule can negtively regulate the signal transduction genes like such as TNF-RΙ, Fas, P53, Casepase-8, and cytochrome c in PD rats'brain.
