Objective:To investigate the pathogenetic mechanism and look for available therapetutics theory for spontaneous abortion by comparing the expressions level of proliferating cell nuclear antigen (PCNA),Ki-67,surviving and OPN in trophoblast and decidua tissue of spontaneous abortion with normal pregnancy mice.Methods: The nonabortion-prone (CBA×BALB/c) matings were established as the models of normal pregnancy and the abortion-prone(CBA×DBA/2) matings were used as the models of spontaneous abortion. Immunohistochemistry method(SP) was employed to detect the expressions of PCNA,Ki-67, surviving and OPN in trophoblast and decidua tissue at the day 13 of gestation. The data was analyzed byχ2 test and Wilcoxon rank sum test.Results:(1) The rate of embryonic resorption of spontaneous abortion mice at day 13 of gestation was significantly higher than that of normal pregnancy mice (P<0.01).(2) There were PCNA protein positive staining in both models at day 13 of gestation. The expression's level of PCNA in trophoblast of spontaneous abortion model was lower than of normal pregnancy model (P<0.01); the expression's level of PCNA in decidua of spontaneous abortion model was lower than of normal pregnancy model ,too (P<0.05);(3) There were Ki-67 protein positive staining in both models at day 13 of gestation. The expression's level of Ki-67 in trophoblast of spontaneous abortion model was lower than of normal pregnancy model (P<0.01); the expression's level of Ki-67 in decidua of spontaneous abortion model was lower than of normal pregnancy model ,too (P<0.01);(4) There were survivin protein positive staining in both models at day 13 of gestation. The expression's level of survivin in trophoblast of spontaneous abortion model was lower than of normal pregnancy model (P<0.01); the expression's level of survivin in decidua of spontaneous abortion model was lower than of normal pregnancy model ,too (P<0.01);(5) There were OPN protein positive staining in both models at day 13 of gestation. The expression's level of OPN in trophoblast of spontaneous abortion model was lower than of normal pregnancy model (P<0.05); the expression's level of OPN in decidua of spontaneous abortion model was lower than of normal pregnancy model ,too (P<0.05).Conclusions: The expressions of PCNA, Ki-67, suvivin and OPN in villi and decidua were associated with the occurrence of spontaneous abortion. The lower expression of PCNA, Ki-67, suvivin and OPN in placental villi and decidua which were caused by certain diseases might be one reason of spontaneous abortion.
