Objective: To discuss the pathogenesis of vascular dementia and to investigate the Effects of electroacupuncture(EA) on the disorder learning and memory in rats with vascular dementia(VD) and explore its molecular mechanism。Methods: Among 40 male SD rats,12 rats were in sham-operation group and the other 38 rats were used to establish models by 2-vessel occlusion(2-VO) method, then survival model rats were randomized into EA group (n=12), model group(n=13). EA group was treated with at points of baihui,dazhui and shenshu, and sham-operation group and model group did not receive any treatmen。After a 30-day treatment, morris water maze was used for place navigation test and spatial probe test, and the expression of p53. Noxa and Caspase-3 in hippocampal CA1 region were measured by immunofluorescence method。Results: 1,In VD group, the expression of P53,Noxa and Caspase-3 in hippocampal CA1 region was higher than that in sham-operation group。2,In VD group, the expression of p53 and Noxa was in significant correlation, and so was the expression of Noxa and Caspase-3。3,In EA group, learning and memory ability was higher than that in In VD group。4,In EA group, the expression of P53, Noxa and Caspase-3 in hippocampal CA1 region was lower than that in VD group。Conclusion: 1,The high expression of p53,Noxa,Caspase-3 in hippocampal CA1 region closely correlates with pathogenesis of vascular dementia。2,Noxa induced mitochondria apoptosis pathway plays an important role on the pathogenesis of vascular dementia, during which Noxa is incentived in p53 dependent and Caspase-3 plays an important role。3,Electroacupuncture can improve learning and memory ability in VD rats, and it can also inhibit apoptosis in hippocampal CA1 region by inhibiting the expression of p53, Noxa and Caspase-3。...
