ObjectiveBy observing the production of interferon-gamma(IFN-γ),interleukin-4(IL-4)in bronchoalveolar lavage fluid (BALF) and spleen mononuclear cell culture supernant, calculating IFN-γ/IL-4,to clarify the changes of Th1/Th2 during the course of asthma. By observing the expression of T-bet in lung, to clarify the changes of T-bet during the course of asthma The influence of BCG on the production of IFN-γ,IL-4 and T-bet was also investigated in this study.MethodKunming mice (male,3 weeks,weight 18-22g)were randomly divided into three groups, each group 20 mice. Each group was divided into four subgroups, each subgroup 5 mice.1)Asthma Group: each mouse was sensitized with injection intrapertoneally 0.01% OVA 0.1ml and 2% aluminum hydroxide 0.1ml,challenged by inhaling aerosol 1% OVA to establish asthma model.2)BCG Group: each mouse was sensitized/challenged with the same protocol used by asthma group, but these mice were treated with injection intracutaneously BCG 0.025mg before OVA sensitized.3)Control Group: each mouse was sensitized/challenged by saline instead of OVA. The amount of IFN-γ,IL-4 in bronchoalveolar lavage fluid (BALF) and spleen mononuclear cell culture supernant was assessed by ELISA at 0,15,23,29d.The expression of T-bet in lung tissue was assessed by Western Blot.Results1. The changes of IFN-γin bronchoalveolar lavage fluid and spleen mononuclear cell culture supernant: IFN-γwas decreased in asthma group compared with control group after OVA sensitized, decreased after first challenge and decreased significantly after multiple challenges; IFN-γin BCG group was increased compared with asthma group after OVA sensitized, increased significantly after first,multiple challenges, but lower than control group.2. The changes of IL-4 in bronchoalveolar lavage fluid and spleen mononuclear cell culture supernant:IL-4 was increased in asthma group compared with control group after sensitized, increased after first challenge and increased significantly after multiple challenges;IL-4 in BCG group was decreased compared with asthma group after OVA sensitized, but higher than control group, decreased significantly after first,multiple challenges, but higher than control group.3. The changes of IFN-γ/IL-4 in bronchoalveolar lavage fluid and spleen mononuclear cell culture supernant:IFN-γ/IL-4 was decreased in asthma group compared with control group after sensitized, decreased significantly after first challenge and multiple challenges;IFN-γ/IL-4 in BCG group was increased compared with asthma group after OVA sensitized, but there was no difference, compared with control group,IFN-γ/IL-4 was increased after first,multiple challenges, but lower than control group.4. The changes of T-bet in lung tissue:T-bet was decreased in asthma group compared with control group after sensitized, decreased after first challenge and multiple challenges too, but among them there was no difference. T-bet in BCG group was increased compared with asthma group after OVA sensitized,first challenge and multiple challenges, but lower than control group.Conclusion1. Th1/Th2 was excursed in sensitization. After challenge, excursion of Th1/Th2 was more significant. BCG can retrieve the excursion of Th1/Th2 in sensitization. And retrieve in part the excursion of Th1/Th2 after antigen challenge. 2. T-bet was decreased in sensitization, decreased after first challenge and multiple challenges too, but among them there was no difference. BCG can retrieve T-bet in sensitization and retrieve in part after antigen challenge.
