Objective:To investigate the distribution of surface sensitization points and the temperature of sensitized points in asthmatic rats,combined with the changes of related tissue mast cells?MCs?and substance P?SP?,to explore the acupoint sensitization point and heat of asthma model rats.The mechanism of the generation of sensitive points.Methods:Sixty male Sprague-Dawley rats weighing?250±20?g were randomly divided into 6 groups:model I and blank I,model II and blank II,model III and blank III.,each group of 10 each.The model group was given a mixture of ovalbumin/aluminum hydroxide?OVA/AL?OH?3?in the peritoneal cavity and limbs,and then injected into the OVA physiological saline solution by ultrasonic atomization to prepare a rat model of asthma.Model I was continuously modeled for 4 weeks,model II group was continuously modeled for 8 weeks,model III group was continuously modeled for 12 weeks,and blanks I,II,and III were injected with normal saline for 4 weeks,8 weeks,and 12 weeks,respectively.Same model group.EB exudation was used as a marker of acupoint sensitization.After the end of the corresponding experimental period,each group was injected with EB into the tail vein under isoflurane inhalation anesthesia.The skin EB exudation point was recorded after depilation and the rat back was detected by infrared thermography.The EB oozes and the temperature of the control point,and finally the corresponding tissue is cut.The amount of MCs in the tissue at the EB exudation point and the degranulation were detected by modified toluidine blue staining.The expression of SP in the tissue at the EB exudation site was detected by immunohistochemical staining.Results:1.Distribution and temperature of sensitization points:The EB exudation points of the model I group are mainly distributed on the left side of the"lungshu",and the EB exudation points of the model II group are mainly distributed on the bilateral"lungshu"and the left side.In the model group III group,the EB exudation points were mainly distributed in the bilateral“feishu”and the bilateral“Dingchuan acupoint”,and the EB exudation point also appeared in the right part of the chest“Huagai”.Under the infrared thermal imager,the high temperature zone is mainly distributed in the neck,the high temperature of the model I group is distributed to the left of the neck,the model II group and the model III group are distributed around the neck;the temperature of the corresponding point in the blank group is not obvious.Variety.Compared with the blank group,the temperature of the EB exudation point in the model group increased significantly?P<0.05?.Compared with the EB exudation point in the model group,the temperature of the EB exudation point in the model group did not increase significantly;Compared with the open point,the temperature of the EB exudation point in the model group increased significantly?P<0.05?.Compared with the open point of the blank group,the temperature of the EB exudation point in the blank group did not change significantly.The mean temperature between the EB exudation points of the three model groups increased slightly with increasing disease duration,but there was no statistical difference.2.The number of MCs and their degranulation and SP expression in sensitized tissue:the number of MCs in the sensitized point of model I group and model II group was significantly higher than that of sensitization point and the corresponding point of blank group?P<0.05?,model III There was no statistically significant difference between the MCs and the sensitized points at the sensitized points,but there was a statistically significant difference between the corresponding points in the blank group?P<0.05?.The degranulation rate of MCs at the sensitization point of the three model groups was significantly higher than that of the sensitization point and the corresponding points of the blank group?P<0.05?.The degranulation rate of MCs at the corresponding point between the sensitization point and the blank group was not obvious.Statistical differences.There was no significant difference in the number of MCs and the rate of degranulation between the three model groups.The positive expressions of SP in the sensitized points of the three model groups were higher than those in the sensitized points and the corresponding points in the blank group?P<0.05?.There was no statistical difference between the SP positive expressions in the vicinity of the sensitized points and the corresponding points in the blank group.The expression of SP at the sensitized points of the three model groups was compared.The positive expression of SP gradually increased with the prolongation of the disease course,but did not reach statistical difference.Conclusion:1.Asthma model rats have sensitized acupoints such as“feishu”,“dingchuan acupoint”and“Huagai acupoint”,and the sensitized point will increase with the prolongation of modeling time;the thermal point of asthma rat model Mainly"feishu"and"Dingchuan Point".2.MCs and SP participated in the sensitization phenomenon of acupoints.The sensitization of MCs involved in acupoints is mainly related to the degranulation.With the development of disease levels,the sensitization of acupoints will appear"diffusion".The number of MCs per unit area and the expression level of SP in the sensitized point were in a state of dynamic equilibrium,and the increase was not obvious with the aggravation of the disease.3.There is a certain degree of coincidence between the sensitized point and the sensitive point of the asthma model rat.
