Objective: The purpose of this study was to investigate the distribution of osteoprotegerin gene polymorphism in the Han population of Sichuan province and to study the association of the polymorphism of OPG gene and coronary heart disease (CHD).Methods: The polymerase chain reaction-restriction fragment polymorphism(PCR-RFLP) analyzing methods were used in the study designed a case-control study. OPG gene T950C and G1181C polymorphism was examined between 150 patients with CHD and 150 unrelated healthy persons. The distribution of the OPG gene polymorphism was compared between the health population and patients with CHD and the association of OPG gene polymorphism and CHD was examined.Results: 1. OPG gene T950C polymorphism was existing in Hans population of Sichuan, the frequency of TC genotype was the highest among OPG genotypes (46%), TT genotype(30%), CC genotype(24%), and these gene polymorphisms had no difference between male and female (P>0.05). There were sigficant difference between Hans population of Shaanxi and Europeans groups. But compared with groups of Japenese and Korean, the distributions of OPG genotype and allele were no significantly different. And OPG gene G1181C polymorphism was existing in Hans population of Sichuan, the frequency of GG genotype was the highest among OPG genotypes (57%), GC genotype(30 %), CC genotype(13%), and these gene polyrnorphisms had no difference between male and female (P>0.05). There were sigficant difference between Hans population of Sichuan and European groups. But compared with groups of Japenese and Korean, the distributions of OPG genotype and allele were no significantly different.2. OPG gene T950C and G1181C single polymorphis were ont assoiated with CAD, but linkage of polymorphisms 950 and 1181 revealed that haplotypes were distributed significantly differently in men with CAD vs. men without CAD with overrepresentation of genotypes 950 TC/1181 GC (OR=1.55) and 950CC/1181 CC (OR=4.2) in patients with CAD. As analyzed by binary logistic regression analysis, the presence of either 950 TC/1181 GC or 950 CC/1181 CC genotype was associated with an independently and significantly increased risk for CAD, with an OR of 1.67 (95% CI= 1.02-2.72)Conclusion: In summary, OPG polymorphisms were existing in Hans population of Sichuan, and its distribution some different among different ethnics. OPG polymorphism have some association with CHD in this study.
