Objective: With development of the research about Carbohydrate Chemistry and Glycobiology in latest years, a great of achievements about the study on the structure and activity of polysaccharides have been made. Polysaccharides exhibit many biological activities, including of anticancer, anti-virus, anti-infection, radiation protection, immunological regulation, promoting biological synthesis of protein and nucleic acid. Tremella fuciformis Berk belongs to tremellaceae of Heterobasidiomycetae, it is known as nutritious mushroom and important medicine by people all over the world. To explore the relationship between structure and bioactivity of polysaccharide, this paper presents the extraction, isolation, identification of polysaccharides from spores of Tremella fuciformis Berk fermentation products。Methods: DEAE-Sephadex A-25 and Sephadex G-200 were adopted to separate and purify the water solution polysaccharides from spores of Tremella fueiformis Berk. Two homogeneous polysaccharides named WTF-A and WTF-B have been obtained and identified. Alkaline solution polysaccharides were obtained by using 0.5M&2M, and DEAE-Sephadex A-25 and Sephadex G-200 were adopted to separate and purify the alkaline solution polysaccharides, other two homogeneous polysaccharides named ATF-A &ATF-B has been obtained and identified. The structure feature of WTF-A, WTF-B and ATF-A&ATF-B were analyzed by chemical methods as whole hydrolyzed, partial hydrolyzed, methylated and spectroscopic methods as UV, IR, GC, GC-MS, NMR. Results: WTF-A is composed of galactose, glucose, mannose and two heptoses。Its molecular weight is estimated to be 73000 by HPLC. Its main chain is comprised of 1, 6 linked galactose and 1, 2, 3, 4, 6 linked mannose or 1, 3, 6 linked mannose. Side chain are comprised of terminal glucose, terminal mannose, 1, 2, 4 linked mannose, L-glycero-D-glucohep-topyranoside and L-glycero-D-mannohepopyranoside.WTF-B is composed of galactose and uronic acid。Its molecular weight is estimated to be 68000 by HPLC. Its main chain is comprised of 1, 6 linked galactose. Side chain are comprised of 1, 2, 3, 4, 6 linked glucose and 1, 3, 6 linked mannose and 1, 3, 7 linked glycero-d-mannoheptitol. Uronic acid may be in main chain because of its 32.88% content.ATF-A is main composed of glucose. Its molecular weight is estimated to be 62830 by HPLC. Its main chain is comprised of 1, 2, 3, 4, 6 linked glucose。Side chain are comprised of 2, 4, 6 linked galactose and 2, 3, 7 linked glycero-d-glucoheptopyranoside。ATF-B is main composed of glucose. Its molecular weight is estimated to be 63710 by HPLC. Its main chain is comprised of 1, 4 linked glucose and 1, 2, 3, 4,6 linked glucose. Side chain are comprised of 1, 6 linked galactose and 1, 3, 7 linked glycero-d-mannoheptitol.Conclusion: WTF-A,WTF-B,ATF-A and ATF-B are new heteropolysaccharides from spores of Tremella Fuciform berk, their chemical structures were identified for the first time.
