Objective: Tremella Polysaccharides (TP) are the major component and activity unit of tremella. This paper is to study on anti-aging effects and their mechanism. Methods: This study was consisted with experiments in vitro and experiments in vivo. P21 gene were integrated into the genome DNA of HELF cells by DNA recombination technique to make cell-aging model in the experiments in vitro. This cell-aging model was interfered by TP. Mice were injected by D-galactose to make subacute aging model in the experiments in vivo. At the same time, these mice were interfered by TP. Results: Experiments in vitro: TP can down-regulate transcription and expression of p21 gene, promote cell proliferation and prevent cell from aging. The best effect dose of TP is 0.5mg/l. Experiments in vivo: TP can down-regulate transcription and expression of p21 gene, have the activity of anti-oxidation, increase contents of hydroxyproline in the skin and strengthen immunity in mice. Conclusion: TP have anti-aging effects by regulating transcription and expression of cell cycle negative regulator p21, anti-oxidation and strengthen immunity.
