Objective: The impersonality of the large intestine wet heat syndrome modelwere discussed by investigating the method to make large intestine damp-heatsyndrome animal model, detecting the content of IL-1,IL-2,IL-6 and rotavirusantigen in blood serum and fecal of model rats; Observing the pathological changeof large intestine constitution of the model rats through optics microscope andelectron microscope; anti-assistant to aid therapy by using dispel wet heat drugGegenqinliantang.Methods: By means of clearing grade Wistar rats and BLAB/C immaturemurine for experiment animals. To establish escherichia coli cast bundle (Ⅰbundle),rotavirus cast bundle (11 bundle ), escherichia coli+rotavirus cast bundle (111bundle), immature murine rotavirus model bundle (Ⅳbundle), normal contrastbundle, Gegenqinliantang therapeutics bundle, therapeutics contrast bundle. Largeintestine wet heat syndrome animal model were created with a comprehensivemethod of fodders rich of sugar and fat circumstance of high wet and temperature, pouring rotavirus and escherichia coli into stomach, Gegenqinliantang were used totreat wet heat syndrome model rats. The contents of IL-1,IL-2,IL-6 in bloodserum were detected with radioimmunity. Rotavirus antigen was detected withELISA. HE staining was used to observe large intestine's pathological changes.Transmission electron microscope was used to observe ultra -micro pathologicalalteration.Results: Large intestine wet heat syndrome model group rats appeared thesesymptoms: febrile, anorexia, anus red swelling and hyperemia, diarrhea, tiredness,hair fluffy etc. Not only the episode condition and the lesion apparatus, but alsomains symptoms and physical signs the model proximity the Chinese medical largeintestine wet heat syndrome. Their contents of serum IL-1,IL-2 were notably higherthan those of wet heat syndrome model group rats (P<0.001).However, there was nodifference in IL-6(P=0.053). Rotavirus antigen in their blood serum and fecal weredetected by ELISA method. Mucous membrane hyperemia, edema, bleeding,lymphoproliferation and inflammatory cell infiltration were seen by HE staining. Itwas thus clear that mass bacillus phagosome (BP) and viral inclusion body (IB) underelectron microscope. The symptoms of Gegenqinliantang treatment group ratsdisappeared after they were treated with Gegenqinliantang, the content of IL-1,IL-2,IL-6 in blood serum resume to the levels near to those of normal control group rats.Rotavirus antigen was extinctied. Light microscope and electron microscopemanifested that bowel inflammatory reaction relieved.Conclusion: (1) fodder rich of sugar and fat, pouring rotavirus and colibacillusinto stomach we succeeded to make the large intestine wet heat syndrome modeladopting totally-enclosed type hyperthermia granary first time, which canself-adjustment temperature and degree of wetness. (2)It make knows that large intestine wet heat syndrome group rats presence inflammatory reaction and immunesystem change according to the increasing of the IL-1,IL-2,IL-6 contents in themodel rats blood serum. The observe results of light and electron microscopeconfirms that the large intestine of the model group rats presence comprehensiveinflammatory reaction. (3) It certificates the model was large intestine wet heatsyndrome model through the symptoms disappeared after the model rats treated byGegenqinliantang, the contents of serum IL-1,IL-2,IL-6 resume normal, rotavirusantigen extinctied, intestine tract inflammation relieved.
