Research On The Model Of Large Intestine Of Wet Heat Syndrome And Study The Effection Of Jixiekang On It

objective:The impersonality of the large intestine wet heat syndrome model were discussed by investigating the method to make large intestine wet heat syndrome animal model,detecting the content of EL-l,IL-2,IL-6 and rotavirus antigen in blood serum and fecal of model rats;Observing the pathological change of large intestine constitution of the model rats through optics microscope and electron microscope, ;anti-assistant to aid therapy by using dispell wet heat drug jixiekang .MethodsrBy means of clearing grade Wister rats and BLAB/C immature murine for experiment animals.To establish escherichia coli cast bundle( I bundle),rotavirus cast bundle( II bundle), escherichia coli+ rotavirus cast bundle (III bundle),immature murine rotavirus model bundle( IV bundle), normal contrast bundle, jixiekang therapeutics bundle,therapeutics contrast bundle.Large intestine wet heat syndrome animal model were created with a comprehensive method of fodders rich of sugar and fat,circumstance of high wet and temperature,pouring rotavirus and escherichia coli into stomachjixiekang were used to treat wet heat syndrome model rats.The contents of EL-l,IL-2,IL-6 in blood serum were detected with radioimmunity.Rotavirus antigen were detected with ELISA.HE staining were used to observe large intestine's pathological changes.Transmission electron microscope were used to observe ultra-micro pathological alteration.ResultsrLarge intestine wet heat syndrome model group rats appeared these symptoms : febrile, anorexia, anus red swelling and hyperemia, diarrhea,tiredness,hair fluffy etc.Not only the episode condition and the lesion apparatus,but also main symptoms and physical signs the model proximity the Chinese medical large intestine wet heat syndrome.Their contents of serum IL-l,IL-2,IL-6 were notably higher than those of wet heat syndrome model group rats.(P<0.01).Rotavirus antigen in their blood serum and fecal were detected by ELISA method. Mucous membrane hyperemia, edema, bleeding, lymphoproliferation and inflammatory cell infiltration were seen by HEstaining.lt was thus clear that mass bacillus phagosome(BP) and viral inclusion body(VEB) under eletron microscope.The symptoms of jixiekang treatment group rats disappeared after they were treated with jixiekang,the content of IL-l,IL-2,IL-6 in blood serum resume to the levels near to those of normal control group rats.Rotavirus antigen were extinctied.Light microscope and electron microscope manifested that bowel inflammatory reaction relieved.Conclusion: (1) fodder rich of sugar and fat,pouring rotavirus and colibacillus into stomath,we succeeded to make the large intestine wet heat syndrome model adopting totally-enclosed type hyperthermia granary first time,which can self-adjustment temperture and degree of wetness.(2) It make knows that large intestine wet heat syndrome group rats presence inflammatory reaction and immune system change according to the increasing of the EL-1,IL-2,IL-6 contents in the model rats blood serum.The observe results of light ang electron microscope confirmates that the large intestine of the model group rats presence comprehensive inflammatory reaction.(3) It certificates the model was large intestine wet heat syndrome model through the symptoms disappeared after the model rats treated by jixiekang,the contents of serum IL-l,IL-2,IL-6 resume normal,rotavirus antigen extinctied,intestine tract inflammation relieved.
