Objective:To investigate the genotype and allele distribution frequence of adiponectin-11377 nucleotide C/G polymorphism in hui population of Qinghai and study the relationship of the polymorphism and insulin resistance as well as the risk of type 2 diabetes.Methods:All subjects are hui nationality in Qinghai including 122 type 2 diabetics(T2DM)and 108 non-diabetic first-degree relatives(NDR)selected from type 2 diabetic pedigrees,and 103 healthy subjects(NC)with normal glucose-tolerance serving as controls.All subjects were genotyped for the polymorphism of adiponectin by PCR-RFLP assay.Plasma glucose,lipids and insulin were determined.Body mass index(BMI)and insulin resistance index(HOMA-IR)were evaluated.Results:1.The frequencies of every genotype and each allelic at locus-11377 in non-diabetic first-degree relatives group were the same as those in control group.The frequency of GG genotype in type 2 diabetic group(12.30%)was higher than those in non-diabetic first-degree relatives(1.86%)and in normal controls(1.94%),p<0.01. The frequencies of CC and CG genotype in type 2 diabetic group were the same as those in non-diabetic first-degree relatives and in normal controls.The frequency of G allele in type 2 diabetic group(34.02%)was higher than those in non-diabetic first-degree relatives(24.07%)and in normal controls(24.76%),P<0.01.The frequency of C allele in type 2 diabetic group was lower than those in non-diabetic first-degree relatives and in normal controls,P<0.01. 2.The BMI of each genotype in normal controls had no significant difference. The BMI of CG+GG genotypes in the type 2 diabetic group(23.6±2.7)and in non-diabetic first-degree relatives group(23.1±3.1)was higher than that of CC genotype(22.1±2.9,22.6±2.8),P<0.05.In normal controls,the frequencies of genotypes of obese subgroup and non-obese subgroup were no significant difference. The frequencies of CG+GG genotypes(65.12%,48.08%)and G allele(40.70%, 25.96%)in obese subgroups of the type 2 diabetic group and non-diabetic first-degree relatives group were higher than those(50.63%,46.64%;30.40%,22.32%)in non-obese subgroups,P<0.05.3.The HOMA-IR of each genotype group was no significant difference in normal controls.The HOMA-IR of CG+GG genotypes(2.35±1.58)in non-diabetic first-degree relatives group was higher than that of CC genotype(2.07±1.28)(P<0.05).4.Stepwise regression showed the HOMA-IR in non- diabetic first-degree relatives group was correlated with FINS,FPG,BMI and genotype(P<0.05),which was still significant after correction with BMI(P<0.05).The HOMA-IR in normal controls was correlated with FINS,FPG,BMI(P<0.05),but not with genotype.Conclusion:Hui population of Q inghai with GG genotype in adiponectin-11377 had high susceptibility of type 2 diabetes.G allele was significantly associated with obesity in type 2 diabetes,obesity and insulin resistance in non- diabetic first-degree relatives.
