ObjectiveWe investigated the contribution of [alpha]1-adrenoceptor mechanisms to urethral dysfunction associated with 2 type diabetes mellitus (DM) in rats.Materials and Methods2 type diabetes mellitus group rats were induced by intraperitoneal injection of streptozotocin (STZ) (90mg/kg) after they were born two days, normal rats were not givern any intervention.2 type diabetes mellitus rats were weaned after they were born four weeks and injected streptozotocin (25mg/kg) intraperitoneally and given food and drink with high sugar and high fat.Normal rats were given ordinary feed.2 type diabetes mellitus rats were induced Successfully.Under urethane anesthesia ,baseline urethral perfusion pressure(UPP), the lowest urethral pressure, the mean rate and amplitude of high frequency oscillations were measured under isovolumetric conditions.ResultsBefore terazosin injection in 2 type diabetes mellitus group rats the lowest urethral pressure (UPP nadir) during urethral relaxation and the intravesical pressure thresholds for inducing urethral relaxation were significantly higher than in normal rats (p<0.01) , while baseline urethral perfusion pressure were not significantly different.The mean rate of high frequency oscillations was also significantly lower than in normal rats (p<0.01) .Intravenous administration of terazosin (0.4 mg/kg), an [alpha]1-adrenoceptor antagonist, significantly decreased the UPP nadir (p<0.01) and baseline UPPs (p<0.01) and the intravesical pressure thresholds for inducing urethral relaxation (p<0.01) in 2 type diabetic rats but not in normal rats,while in the 2 groups of animals the mean rate and amplitude of high frequency oscillations were not significantly changed .Conclusion2 type diabetes mellitus group rats showed smooth and striated muscle dysfunctions of the urethra. The inhibition of [alpha]1adrenoceptors, which decreased significantly the UPP nadir and basline UPP, may be useful for treating urethral dysfunction.
