Background and Object: Intrcerebral Hemorrhage (ICH) is acommon acute cerebrovascular disease, belonging to the category ofstroke in Traditional Chinese Medicine (TCM). It brings heavy burden tosociety, with high incidence,death and mutilation rate.Since the view ofQixu Xueyu posed by a famous doctor of Qing dynasty, Wang Qingren,who initially invented the Buyang Huanwu Decoction, Yi-QiHuo-Xue(YQHX) therapy has been widely used in clinic.The modernresearches have found that YQHX therapy can improve the blood flowand energy metabolism in the brain hemorrhage zone,rectify theabnormal blood rheology changes in brain,inhibit inflammatory reactionin the brain hemorrhage zone and so on. However,the vessel system ofthe hematoma was damaged. Only through the reconstruction of capillaryvessel system in the focal zone, can the perfusion in the hematoma zonebe restituted,the microenvironment of the focus be improved and therepair of nervous system be accelerated.As far,vascular endothelialgrowth factor(VEGF) was the only special growth factor proved toangiogenesis.By binding to its receptors of VEGFR-1(Flt-1) and VEGFR-2(Flk-1) in the vascular endothelial cells,VEGF could start the bud,theoutset of angiogenesis,and at last there would be a grown vessel so as toreconstruct the capillary vessel system in the focal zone,and improve torestitute the reperfusion in the hematoma zone,accelerarate the absorptionof hematoma,recover the blood and oxygen utilized by neuron andpromote the neurofunctional rehabilitation. By observing the effects ofYQHX therapy on expression of VEGFmRNA and its receptorsFlt-1mRNAandFlk-1mRNA in intracerebral hemorrhagic rats. So as tostudy the mechanism of YQHX therapy on reconstitution of capillary vessel system in the hematoma area after ICH initially.Methods: 155 SD rats were randomly divided into six groupsincluding normal control group(n=5),sham control group(n=30),ICHmodel control group(n-30),Yiqi-treated group(n=30),HuoXue-treatedgroup(n=30) and YQHX-treated group (n=30).By injecting collagenasetypeⅦstereotaxicallyin to right globus, ICH model was made.Yiqi,HuoXue and YQHX-treated group were respectively administered atBuyang Huan wu decoction,YiQi decoction and HuoXue decoction.Every rat was caculated on forelimb use asymmetry brfore it wasexecuted;The expression of mRNA of VEGF,Flt-1 and Flk-1 was assayedby reverse transcription-polymerase chain reaction (RT-PCR) at dayl4,7,14,21 and 28 after the onset.Results:1.The information of forelimb use asymmetry: hemiparalysissymptoms were seen after ICH rapidly. At 4 day,the ratio of forelimb useasymmetery descended notably (p<0.05),but was higher than that shamcontrol group.At 14day,the ratio in Yiqi Huoxue group rats became to belower than that in model group rats.So these information hinted that YiqiHuoXue therapy can decreased the ratio of forelimb use asymmetry of therats with ICH.2.Nonsignificant expression of VEGFmRNA, Flt-1mRNAand Flk-1mRNA in rat brains of normal or sham group during theexperiment;In model group,the expressions of VEGFmRNA,Flt-1mRNAand Flk-1mRNA appeared at 1d,peaked at 21d,and then weakenedgradually;The expressions of VEGFmRNA,Flt-1mRNA and Flk-1mRNAin YQHX-treated group were significantly higher than those in the othergroups from 7d to 21d (P<0.01), and the expression of VEGFmRNA andFlk-1mRNA in YQHX-treated group reached peak at 14d,but theexpression of Flt-1mRNA reached peak at 21d (P<0.01).Conclusion: 1. With the time during in the observation, there is a tendency of theexpression of VEGFmRNA, Flt-1mRAN and Flk-1mRNA in the ratbrain hemorrhage zone after ICH.2. YQHX therapy has a positive effect on the promotion to theexpression of VEGFmRNA, Flt-1mRAN and Flk-1mRNA in the ratbrain of ICH,YQHX therapy may promote angiogenesis, acceleratethe absorption of hematoma and promote the neurofunctionalrecovery of ICH rat.
