ObjectiveTo examine the expression of CD147, cyclophilin A and cyclophilin B in psoriatic lesions, and investigate their roles in the pathology of psoriasis and its clinical significance.MethodsFormalin-fixed paraffin-embedded skin biopsy specimen blocks from psoriatic patients and normal controls were used in this study, including 15 cases of pustular psoriasis ( PP ), 20 cases of psoriasis vulgaris in progressive stage ( PVP ), 20 cases of psoriasis vulgaris in static stage ( PVS ), and 17 specimens of normal skin ( NS ). Immunohistochemical method was carried out to semi-quantitatively analyze the expressions of CD147, cyclophilin A and cyclophilin B in keratinocyte, T cells and neutrophils of all psoriatic lesions and in the keratinocyte of normal skin. An immunoreactivity intensity distribution index ( IRIDI ) was calculated to express the proportion of the immunoreactive cells as well as the staining intensity in those cells.Results1. CD147, cyclophilin A and cyclophilin B staining were observed in keratinocyte, T cells and neutrophils of all psoriatic lesions and in the keratinocyte of normal skin.2. Compared with PVP, the mean IRIDI scores of CD147 and cyclophilin A expression in the keratinocyte, T cells and neutrophils with PP are stronger, respectively ( all P < 0.05 ). The scores of CD147 and cyclophilin A expression in the T cells and neutrophils with PVP were significantly higher than those of PVS, respectively ( all P < 0.05 ). As compared to NS, the scores of CD147 and cyclophilin A expression in the keratinocyte with PP, PVP or PVS showed statistically significant up-regulation, respectively ( all P < 0.05 ).Compared with PVS or PVP, the scores of CD147 and cyclophilin A expression in keratinocyte with PP showed significant stronger ( all P < 0.05 ). The scores of CD147 and cyclophilin A expression in the keratinocyte of patients between PVS and PVP have no statistical difference ( both P > 0.05 ).3. The mean IRIDI scores of cyclophilin B expression in T cells with PP were significantly higher than those of both PVP or PVS ( both P < 0.05 ). Compared with PVS, the scores of cyclophilin B expression in T cells with PVP was significantly higher ( P < 0.05 ). The scores of cyclophilin B expression in keratinocyte and neutrophil are not significant difference in each group collected for the experiment ( all P < 0.05 ).Conclusion1. The elevated expression of CD147 and cyclophilin A in keratinocyte of psoriasis could be associated with keratinocyte hyperproliferation, the disease progression and classification of psoriasis.2. The elevated expression of CD147, cyclophilin A and cyclophilin B in T cell could be associated with the disease severity and classification of psoriasis.3. The elevated expression of CD147 and cyclophilin A in neutrophil of psoriasis could be associated with the disease progression and classification of psoriasis.4. CD147, cyclophilin A and cyclophilin B might play important roles in the process of information for psoriatic lesions and the disease genesis and progression of psoriasis.
