| The relation between genotype of HBV and clinical situations ofthe cases with hepatitis B virus infection[Objective] To determine the genotypes of hepatitis B virus (HBV) in the222 patients with hepatitis B virus infection by polymerase chain reactionof sequence specific primers(SSP-PCR), and to explore the relation-shipbetween HBV genotype and clinical situation of the cases with hepatitisB virus infection.[Method] Ten outer and inner primers were designed on the basis of theconserved area of nucleotide sequences in regions of the Pre-S1 throughS genes, in which 8 inner primers had HBV genotype specificity and weredevided into mix A and B to amplify HBV of genotype A, B, C and D, E, Frespectively. Genotypes of HBV were determined directly by the size ofPCR products. To test its reliability and veracity, select some case torepeated experiments. Then to analyze the relationship between genotypesof HBV with native place, sexuality, ago, situation of serology, lever ofALT and TBIL, virus load, and patients' situations.[Result] The data showed the SSP-PCR is a good method to determind genotypeof HBV virus, and it has perfect accuratissime. It can be used to determindgenotype B and C of HBV virus well. Of the 220 HBV infected cases, 72 (32.4%)cases were genotype B , 123(55.4%) cases were genotype C, 1 (0.5%) caseswas genotype D, 7 (3.2%) cases were genotype B and C mixed, 19 (8.6%)cases were genotype indefiniteness. The composition of all kinds of genotypeswere no statistical difference among different native place, sexuality,ago, situation of serology, lever of ALT and TBIL, virus load. The percentof severe-symptomatic appearance in the patients with genotype C HBVinfection is clearly higher than that in the patients with slight-symptomatic appearance (P<0.05) . Especially between chronichepatitis patients with mild manifestations and chronic hepatitispatients with severe manifestations(P<0.01) .[Conclusion] Sequence specific primers PCR could help to determine HBVgenotypes clearly and directly with reliable and accurate results, can beused to investigate in patients infected HBV . The predominant HBVgenotypes were confirmed to be genotype B and C in the determinded 222cases with hepatitis B virus infection. The most patiets are infected withHBV of genotype C. Genotype of HBV correlated with clinical situationsof hepatitis B patients, and HBV of genotype C might cause more severetype of hepatitis B than genotype B . |
