Objective: To investigate the genotype and allele frenquencies of 5 single nucleotidepolymorphisms(SNPs) in the exon 3 of the adiopentin gene in Han and Koreanpopulation of Yanbian area in China, and the relationship to type 2 diabetes mellitus.Methods: Polymerase chain reaction-single strand conformationalpolymorphism(PCR-SSCP) was used to determine whether polymorphisms exists.Then, exact variation was detected by automated DNA direct sequencing, thepolymorphisms of R221S and H241P in the coding exon 3 and SNP+1711C>T(rs4068),+1795A>G(rs1501298),+1833C>A(rs4686440)in the no-codingexon 3 (3'-UTR)of adiponectin gene. Case-contrast analysis was used to study theassociation between adiponectin gene polymorphism in 98 Chinese subjects and 97Korean subjects with normal glucose tolerant (NGT) group and in 92 Chinesesubjects and 90 Korean subjects with type 2 diabetes mellitus (T2DM) group.Results: 1. The R221S polymorphism of adiponectin gene was detected in one of 98Han people with NGT group and in one of 90 Koren people with T2DM group, theA allele frequency of NGT group and T2DM group was 1%. The R221Spolymorphism was not detected in Han population with T2DM group or in Korenpopulation with NGT group. 2. The H241P polymorphism of adiponectin gene wasdetected in one of 92 Han people with T2DM group, the C allele frequency ofT2DM group was 1%. The H241P polymorphism was not detected in Korenpopulation with T2DM group or in Koren and Han population with NGT group. 3.The+1711C>T,+1795A>G,+1833C>A polymorphisms of adiponectin gene(3'-UTR) were not detected in the 377 subjects. 4. In the 377 subjects, from theclinical characteristics, results are that BMI,TG and TC in Han and Koreanpopulation with T2DM group are higher than the NGT group; WHR in Hanpopulation with T2DM group is significantly higher than the NGT group; LDL-C inKorean population with T2DM group is higher than the NGT group. Conclusion: There is no difference in frequencies distribution of adiponectin gene R221S andH241P polymorphism between Hart and Korean population, and the NGT group andthe T2DM group of Yanbian area in China.; It can be concluded that the R221S andH241P polymorphisms of adiponectin gene may not the susceptive gene with T2DMin Han or Korean population of Yanbian area in China.
