Objective The purpose of this study was to investigate the change of cTnI,TNF-α, NF-κB of liver in the septic rat and to investigate the interventional effects of ketamine and midazolam.Methods Male Sprague-Dawley rats were randomly divided into group C,the control group,with non-CLP(rats treated with cecal ligation and perforation to evoke peritonitis);group CLP; group KTⅠ;group KTⅡ;group Mi;group KTⅠ+Mi.Cecal was ligated and perforated except in group C.Rats were administrated ketamine5 mg·kg-1·h-1in group KTⅠand 10 mg·kg-1·h-1in KTⅡ, midazolam0.075 mg·h-1after 0.15 mg in group Mi,ketamine5 mg·kg-1·h-1+ midazolam 0.075 mg·h-1 after 0.15 mg in group KTⅠ+Mi through caudal vein after CLP 2 hours,respectively.Then the rats were killed at different time points,and the level of serum cTnI,TNF-α,the NF-κB of liver were measured.Results The expression of cTnI increased significantly at 5h,9h,12h,18h after CLP as compared with control group(P<0.05 or P<0.01)and arrived to its top at 12h.The expression of TNF-αincreased significantly at 2h,5h,9h,12h after CLP as compared with control group(P<0.01)and there was an decreasing trend with development of CLP.The expression of NF-κB liver increased significantly in each time as compared with control group (P<0.01),and there was an increasing trend with development of CLP.The expression of serum cTnI,TNF-αand NF-κB of liver decreased significantly in group C,KTⅠ,KTⅡ,Mi and KTI+Mi as compared with group CLP(P<0.05).The expression of serum cTnI and NF-κB of liver decreased significantly in group KTⅡcompared with which in group KTⅠ,Mi and KTI+Mi(P<0.05).The expression of serum TNF-αdecreased significantly in group KTⅡcompared with which in group KTⅠ,M(P<0.01).There was no difference of serum cTnI,TNF-αand NF-κB of liver within group KTⅠ,Mi and KTⅠ+Mi(P>0.05).Conclusion There exists cardiac injury and liver injury in septic rats,and ketamine and midazolam can ease the injury.This phenomenon can be explained by decreasing the serum TNF-αand NF-κB of liver.There is a relation between protecting effect and dose of ketamine. There is no cooperativity between ketamine and midazolam in protection of heartand liver issue.
