Objective The purpose of this study was to investigate the change of cTnI, TNF-α, NF-κB of myocardial in the septic rat and to investigate the interventional effects of ketamine and metoprolol.Methods (1)Male Sprague-Dawley rats were randomly divided into group C, (the control group, group CLP (rats treated with cecal ligation and perforation to evoke peritonitis) . Then the rats were sacrificed at CLP2h,5h,9h,12h,18h,and the level of serum cTnI, TNF-α, the NF-κB of myocardial were measured . (2) On the base of part 1,male Sprague-Dawley rats were randomly divided into group C, group CLP,group KTI; group KTII; group M and group KTI+M. Cecal was ligated and perforated except in group C. Rats were administrated ketamine5㎎·㎏-1·h-1 in group KTI and 10㎎·㎏-1·h-1 in KTII, metoprolol 0.2㎎·㎏-1·h-1 after 0.5㎎ in group M, ketamine5㎎·㎏-1·h-1+ metoprolol 0.2㎎·㎏-1·h-1 after 0.5㎎ in group KTI+M through caudal vein after CLP 2h, respectively. Then the rats were sacrificed at CLP 5h,and the level of serum cTnI, TNF-α, the NF-κB of myocardial were measured .Results The expression of cTnI increased significantly at 5h,9h,12h,18h after CLP as compared with control group (P﹤0.05 or P﹤0.01)and arrived to its top at 12h. The expression of TNF-αincreased significantly at 2h,5h,9h,12h after CLP as compared with control group (P﹤0.01)and there was an decreasing trend with development of CLP. The expression of NF-κB myocardial increased significantly in each time as compared with control group(P﹤0.01), and there was an increasing trend with development of CLP.The expression of serum cTnI, TNF-αand NF-κB of myocardial decreased significantly in group C,KTI, KTII , M and KTI+M as compared with group CLP(P﹤0.05 or P﹤0.01). The expression of serum cTnI and NF-κB of myocardial decreased significantly in group KTII compared with which in group KTI,M and KTI+M(P﹤0.05 or P﹤0.01) . The expression of serum TNF-αdecreased significantly in group KTII compared with which in group KTI,M(P﹤0.01).There was no difference of serum cTnI, TNF-αand NF-κB of myocardial within group KTI , M and KTI+M (P>0.05).
