Objective:(1) to investigate the association of two SNPs of NRG-1 gene :rs3924999(Arg38Gln) and rs35753505(SNP8NRG221533) and another two SNPs of BDNF gene: rs6265(Va166Met), rs11030101 (-633T/A) with cognitive function of schizophrenia patients. Both of the candidate genes are related to the neurodevelopment of brain.(2) to investigate whether there is deficiency in cognitive function of schizophrenia patients compared with normals(3) to investigate the influence of age, duration of disease and education level on the cognitive function of schizophrenia patients.Method:(1) collect the general and clinical data of 100 patients and the general data of 50 normal controls, then all subjects are required to complete a set ofneuropsychological tests.(2) detect the genotype of rs3924999, rs35753505, rs6265 and rs 11030101 with polymerase chain reaction and ligand detection reaction. (3) employ t test to compare the difference between performances of neuropsychological tests of patients and normal controls. Divide the patients into 2 or 3 groups by genotypes, examine the relationship between each SNP and performances of neuropsychological tests with ANOVA. Compare group-to-group differences with Post hoc test. If there is positive findings, take genotye, age, duration of disease and education level into account to establish a regression equation, to obtain the effect size of the genotype to performances.Results:(1) there is significant difference between patients and normal contrls in almost all of their performances of neuropsychological tests.(2) there is no significant difference among the performances of neuropsychological tests in the 3 groups divided by rs3924999 genotypes, also no significant differences were found in group-to-group comparisons.(3) there is no significant difference among the performances of neuropsychological tests in the 3 groups divided by rs6265 genotypes, also no significant differences were found in group-to-group comparisons.(4) there is significant difference in the performance of comprehensive memory test in the 3 groups divided by rs35753505 genotypes, but in the other tests no significant differences were found. The performance of comprehensive memory test in the T/T genotype group is worse than the other 2 groups. After regression analysis, the genotype and education levels are both related to comprehensive memory test.(5) there is no significant difference among the performances of neuropsychological tests in the 2 groups divided by rs11030101 genotypes.Conclusion:(1) The cognitve function of schizophrenia patients is deficient.(2) The rs3924999 of NRG-1 gene and rs6265 and rs11030101 of BDNF gene are not associated with cognitive function of schizophrenia patients in this study.(3) The polymorphism rs35753505 of NRG1 gene has association with the abilities of comprehensive and episodic memory of schizophrenia patients.(4) The test for the ability of comprehensive memory and episodic memory may be an endophenotype for schizophrenia.
